Girdin、Bim蛋白在儿童肾母细胞瘤中的表达及其与临床病理的相关性

Expression of Girdin and Bim protein in nephroblastoma of children and its correlation with clinicopathologic parameters

目的探讨Girdin、Bim蛋白在儿童肾母细胞瘤和正常组织中的表达及其与临床病理因素的关系,并进一步分析两者之间的相关性。方法收集2000年1月至2009年12月60例住院患者的肾母细胞瘤及肾母细胞瘤旁正常组织,应用免疫组织化学(En VisionTM)法分别检测Girdin、Bim蛋白在肾母细胞瘤和正常组织中的表达情况,数据经统计软件SPSS17.0分析检验。结果 Girdin蛋白在肾母细胞瘤、正常组织中的阳性表达率分别为68.33%(41/60)、46.67%(28/60),两组之间表达差异有显著统计学意义(χ2=5.76,P<0.01);Bim蛋白在肾母细胞瘤、正常组织中的阳性表达率分别为21.67%(13/60)、78.33%(47/60),两组之间表达差异有显著统计学意义(χ2=38.53,P<0.01);肾母细胞瘤组织中Girdin蛋白的表达与肿瘤直径、组织学形态、临床分期、淋巴结转移有关(P<0.05),但与性别、年龄、肿瘤位置无关(P>0.05);肾母细胞瘤组织中Bim蛋白的表达与肿瘤直径有关(P<0.05),但与性别、年龄、肿瘤位置、组织学形态、临床分期、淋巴结转移无关(P>0.05);肾母细胞瘤组织中Girdin蛋白的表达与Bim蛋白表达呈负相关(r=-0.59,P<0.01)。结论肾母细胞瘤组织中Girdin、Bim蛋白的异常表达均与肿瘤直径有关,这表明检测两者有利于判断肿瘤细胞的增殖能力。

Objective To investigate the expression of Girdin and Bim protein in nephroblastoma tissue and normal tissue, and to analyze its relationship with clinicopathologic parameters. Methods The expression of Girdin and Bim protein was respectively detected by immunohistochemisty(EnvisionTM) in nephroblastoma tissue and adjacent normal tissue collected from 60 children with nephroblastoma treated in our hospital from January 2000 to December2009. The data was analyzed by SPSS17.0. Results The positive expression rate of Girdin protein in nephroblastoma tissue and normal tissue was respectively 68.33%%(41/60), 46.67%(28/60), with statistically significant difference(χ2=5.76, P<0.01). The positive expression rate of Bim protein in nephroblastoma tissue and normal tissue was respectively21.67%%(13/60), 78.33%(47/60), with statistically significant difference( χ2=38.53, P<0.01). The positive expression of Girdin protein in nephroblastoma tissue was related to tumor diameter, histological morphous, clinical stage, and lymphaden metastasis(P<0.05), but not to gender, age and tumor site(P>0.05). The positive expression of Bim protein in nephroblastoma tissue was related to tumor diameter(P<0.05), but not to gender, age and tumor site, histological morphous, clinical stages, lymphaden metastasis(P>0.05). The expression of Girdin protein was negatively correlated with the expression of Bim protein in nephroblastoma tissue(r=-0.59, P<0.01). Conclusion The abnormal expression of Girdin and Bim protein in nephroblastoma tissue of children is related to tumor diameter, which indicates that testing both is useful to judging reproductive activity of tumor cell.
