摘要
目的探讨涎腺腺样囊性癌微血管参数与肿瘤微血管模式之间的相关性。方法收集2005年1月至2015年7月我院涎腺腺样囊性癌患者(SACC组)148例,正常腮腺组织(对照组)50例,采用免疫组化染色方法检测两组受检者的转录活化因子-3(STAT3)、血管内皮生长因子(VEGF)m RNA及蛋白表达水平,分析其与微血管密度(MVD)之间的相关性。结果 SACC组患者的STAT3和VEGF阳性表达率分别为32.0%和40.0%,均明显高于对照组的19.0%和18.0%,差异均具有统计学意义(P<0.05);SACC组患者的STAT3、VEGF m RNA表达水平分别为(1.12±0.56)和(1.10±0.29),均明显高于对照组的(0.82±0.31)和(0.72±0.26),差异均具有统计学意义(P<0.05);SACC组患者的STAT3、VEGF蛋白表达(3.23±1.99、2.56±1.86)与对照组(1.96±0.86、1.99±0.86)比较差异具有统计学意义(P<0.05);STAT3及VEGF在SACC组中不同类型组织、有无淋巴结转移、TNM分期方面比较差异均具有统计学意义(P<0.05);STAT3与VEGF表达呈正相关(r=0.85);STAT3及VEGF蛋白表达水平与MVD差异表达具有相关性(r=0.63,0.85)。结论涎腺腺样囊性癌微血管参数STAT3与VEGF与肿瘤微血管模式MVD之间具有相关性,在调控涎腺腺样囊性癌的血管生成方面,有望作为治疗靶点。
Objective To investigate the correlation between salivary microvascular parameters and microvessel modes. Methods One hundred and forty-eight patients with adenoid cystic carcinoma(SACC group) in our hospital and50 subjects with normal parotid tissue(control group) were collected from January 2005 to July 2015. Activator of transcription 3(STAT3), vascular endothelial growth factor(VEGF) m RNA and protein levels were detected by immunohistochemical staining in the two groups, and their correlations with microvessel density(MVD) were analyzed. Results STAT3 and VEGF positive expression rates in SACC group were 32.0% and 40.0%, respectively, which were significantly higher than 19.0% and 18.0% in the control group(P<0.05). The STAT3, VEGF m RNA levels were(1.12±0.56) and(1.10±0.29) in SACC group, significantly higher than those in the control group of(0.82±0.31) and(0.72±0.26), with P<0.05. The two groups had statistically significant difference in STAT3 and VEGF(P<0.05) with(3.23 ± 1.99) vs(1.96±0.86) and(2.56±1.86) vs(1.99±0.86). In SACC group, STAT3 and VEGF levels showed statistically significant difference between different types of tissue, with or without lymph node metastasis, and TNM staging(P<0.05). STAT3 and VEGF expression were positively correlated(r=0.85). STAT3 and VEGF protein expression were correlated with the differential expression of MVD(r=0.63, 0.85). Conclusion There is a correlation between salivary microvascular parameters(STAT3 and VEGF) and microvessel mode MVD, which are expected to be as therapeutic targets in the regulation of angiogenesis in adenoid cystic carcinoma.
