摘要 目的观察乙型肝炎病毒(HBV)对SOX4表达的影响,并探讨其机制。方法 RT-PCR和Western印迹法检测Hep G2和Hep G2.2.15细胞中SOX4 mRNA和蛋白表达的差异,将HBV感染性克隆p HBV1.3及其空载体分别与含SOX4基因启动子的报告质粒p GL3-SOX4共转染Hep G2细胞,测定荧光色素酶的活性,RT-PCR和Western印迹法分别检测SOX4 mRNA和蛋白表达的情况。结果 Hep G2.2.15细胞中SOX4 mRNA和蛋白的表达水平明显高于Hep G2,HBV能够激活SOX4基因启动子的活性,并在mRNA和蛋白水平上调SOX4的表达。结论 HBV能够在转录和翻译水平上调SOX4的表达。 Objective To investigate the effect of hepatitis B virus(HBV) on the expression of SOX4 and its regulatory mechanism. Methods The expression of SOX4 mRNA and protein in Hep G2 and Hep G2.2.15 cells was measured by RT-PCR and Western blot. HBV infectious clone p HBV1.3 and its empty vector were co-transfected into Hep G2 cells with reporter plasmid p GL3-SOX4 containing SOX4 gene promoter. The luciferase activity was measured,and expression of SOX4 mRNA and protein were measured by RT-PCR and Western blot. Results The expression of SOX4 mRNA and protein was higher in Hep G2.2.15 cells than in Hep G2 cells. HBV could activate the activity of SOX4 promoter, and the expression of SOX4 were upregulated at mRNA and protein level. Conclusion HBV can upregulate the expression of SOX4 both at the transcription and translation level.
