摘要
采用碳化二亚胺-N-羟基琥珀酰亚胺(EDC-NHS)法将莱克多巴胺(RAC)与活化的牛血清白蛋白(BSA)偶联,合成了免疫抗原(RAC-BSA),经聚丙烯酰胺凝胶电泳法和紫外吸收法鉴定偶联成功。用合成的免疫抗原免疫家兔获得了高效价的多克隆抗体,采用所得抗体和辣根过氧化酶标抗原建立莱克多巴胺直接竞争酶联免疫吸附分析法(ELISA),标准曲线呈典型S型,具有良好的线性关系。
Ractopamine(RAC) was conjugated to the activated bovine serum albumin(BSA) by means of EDC-NHS method.SDS-PAGE and UV scanning spectrum proved that the hapten ractopamine was coupled successfully to the carrier protein.High titer polyclonal antibodies against RAC were obtained from the rabbits with immune antigen.The direct competitive ELISA of RAC was established with antibodies and horseradish peroxidase-labelled antigen.The standard curve showed the typical sigmoid curve and had good linear relationships.
