摘要
采用Phenomenex(Luna)C18(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-0.05%磷酸(10∶90,V/V),检测波长为280 nm,流速0.8 mL/min,柱温30℃,建立了瑶族藤茶(Ampelosis grossedentata)中没食子酸的RP-HPLC定量分析方法。结果表明,没食子酸进样量在0.057 4~0.344 4μg范围内与峰面积线性关系良好(r=0.999 9),平均回收率为99.15%(RSD=1.80%)。本法简便快捷,可用于瑶族藤茶中没食子酸的含量测定。
The determination of gallic acid in Ampelosis grossedentata by RP-HPLC was studied.The sample was separated by Phenomenex(Luna) C18 column(250 mm×4.6 mm,5 μm) with the mobile phase of methanol-water(containing 0.05% phosphoric acid)(10∶90,V/V) at 30 ℃,and detected at 280 nm with the flow rate of 0.8 mL/min.The results showed that the linear range of gallic acid was 0.057 4~0.344 4 μg(r=0.999 9) and the average recovery was 99.15%(RSD=1.80%,n=9).The established method was accurate,rapid and suitable for detecting gallic acid in A.grossedentata.
