摘要
根据RNAi表达载体的设计原则,以TWH基因为靶基因,将长度253 bp目的片段通过正反两个方向插入表达载体pR1301中,构建RNAi表达载体pR1301-TWH。通过根癌农杆菌EHA105介导法将其导入水稻品种武育粳3号,获得34株阳性转基因植株,为进一步深入研究TWH基因功能奠定了基础。
According to the principles of RNAi vector construction,a 253 bp fragment targeting at TWH gene was inserted into the expression vector pR1301 in both forward and reverse directions;and RNAi expression vector pR1301-TWH was constructed.Through the method of Agrobacterium-mediated transformation,the expression vector was used to transform japonica rice cultivar Wuyujing 3;and 34 positive transgenic plants were gained.This result will provide a good background to study the function of TWH gene.
