摘要
本研究建立了绿茶中花青素的超高效液相色谱分析方法。样品经过乙醇盐酸水溶液提取,沸水浴将花色苷酸水解为花青素单体。采用BEH C18柱(2.1 mm×100 mm,1.7μm)进行分离,二极管陈列检测器检测,外标法定量。结果表明,6种花青素线性相关系数为0.999 6~0.999 9,回收率为78.5%~95.9%,相对标准偏差为1.5%~8.2%。该方法快速准确,灵敏度高、重现性好,适用于绿茶中花青素单体的定性和定量检测。
In this research,a ultra performance liquid chromatographic method was estabolished for the quantification of anthocyanidins in green tea.After extracted under the ultrasonic irradiation in HCl aqueous solution,the anthocyanins were directly hydrolyzed into anthocyanidins.The separation column was Acquity UPLC BEH C18(100 mm×2.1 mm,1.7μm),the sample was detected by PDA detector and quantitative analysis by external standard method.The result showed that the correlation coefficients were 0.999 6~0.999 9.The average recoveries were in the range of 78.5%~95.9%.The relative standard deviations were between 1.5%and 8.2%.The method was simple,sensitive and accurate for the determination of anthocyanidins content in green tea.
