抗纤灵对慢性肾功能衰竭小鼠模型肾组织相关炎症因子的干预作用 被引量:2

Effects of Kangxianling Decoction on Kidney Tissue Inflammatory Cytokines in Chronic Renal Failure Mice

摘要 目的:观察抗纤灵对小鼠慢性肾功能衰竭(CRF)模型肾组织相关炎症因子的干预作用。方法:将40只C57BL/6J小鼠随机分为假手术组、模型组、雷帕霉素组和抗纤灵组,每组10只,除假手术组仅分离肾组织外,其他各组采用Platt 5/6肾切除方法制备CRF模型,雷帕霉素组、抗纤灵组分别予雷帕霉素、抗纤灵水煎剂,假手术组及模型组予等量生理盐水灌胃,疗程8周。免疫组化法检测肾组织中F4/80细胞的表达,RT-PCR法检测肾组织基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)mRNA的表达,ELISA法检测大鼠肾组织白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平。结果:与假手术组比较,模型组小鼠肾组织F4/80阳性细胞,MMP-2、MMP-9mRNA水平明显升高,IL-6、TNF-α含量明显增加(P<0.01);与模型组比较,雷帕霉素、抗纤灵组F4/80阳性细胞减少,MMP-2、MMP-9mRNA水平下降,IL-6、TNF-α含量降低(P<0.05或P<0.01);雷帕霉素和抗纤灵组比较,两组无显著性差异(P>0.05)。结论:抗纤灵能够减轻CRF小鼠肾组织的炎症反应,抑制肾脏纤维化。 Objective:To observe the effects of Kangxianling Decoction(KXLD)on kidney tissue inflammatory cytokines in chronic renal failure(CRF)mice.Methods:Forty 57BL/6Jmice were randomly divided into sham group,model group,rapamycin(RAP)group and KXLD group as 10 mice in each group.Except sham group,other groups were established CRF by Platt 5/6nephrectomy.Then,equal volume of rapamycin(0.13mg/100g),KXLD(2g/100g)and saline was respectively given to mice in the RAP group,KXLD group,normal group and model group,for 8weeks.The mice were sacrificed and renal tissue was collected to observe the F4/80 positive cells by immunohistochemical,the level of MMP-2and MMP-9mRNA expression by RT-PCR and thecontents of kidney tissue inflammatory cytokines IL-6and TNF-αby ELISA were detected respectively.Results:the number of positive F4/80 cells,the level of MMP-2,MMP-9mRNA expression and production of IL-6and TNF-αin model group increased significantly compared with the sham group(P<0.01).However,compared with the model group,the number of positive F4/80 cells,the level of MMP-2and MMP-9mRNA expression and secretion of IL-6and TNF-αobviously decreased in RAP and KXLD group(P<0.05 or P<0.01).While there was no statistical difference between RAP and KXLD groups(P>0.05).Conclusion:KXLD,similar to RAP,may reduce kidney tissue inflammatory reaction in chronic renal failure mice and inhibit renal fibrosis.
