摘要
目的:观察二甲双胍对肥胖大鼠睾丸生精功能的影响。方法:将性成熟的Wistar大鼠分成标准饮食组即对照组、高脂饮食组(HFD组)及高脂饮食+二甲双胍组(Met组)。分析各组大鼠的体重、糖脂代谢的变化,并从睾丸重量、组织学、性激素、炎症状态及精液分析等角度评估肥胖对睾丸生精功能的影响及二甲双胍干预后的效果。结果:饲养8周后,HFD组大鼠体重、体重增加值显著高于对照组和Met组。伴随体重的增加,HFD组大鼠的空腹血糖和胰岛素水平、HOMA-IR指数和瘦素水平显著升高,而睾丸重量、精子质量(密度、活力、形态)则显著降低。血清和睾丸局部雄激素的水平在HFD组明显低于对照组和Met组的,但促炎因子TNF-α、IL-6的表达显著升高。睾丸组织学改变表现为HFD组生精细胞的数量、曲细精管直径显著低于对照组和Met组。结论:二甲双胍可以从改善肥胖带来的糖脂代谢、性激素紊乱、机体及睾丸局部高炎症反应等多个层面提高肥胖大鼠睾丸生精功能。
Objective:To investigate the influence of metformin on spermatogenesis in obese rats.Methods:Wistar rats were randomly divided into three groups:the control group,high-fat diet group and Metformin group.They were fed for 8 weeks with standard chow,high-fat diet,or high-fat diet with metformin.We obtained and analyzed the weight,glucose and lipid metabolism.The influence of obesity on spermatogenesis were explored in terms of testicular weight,semen parameters,histological changes in testis,hormone levels and inflammatory response in serum and testis,etc.The protective effect of metformin was also investigated from the above aspects.Results:Compared with Control group and Metformin group,HFD group exhibited higher body weight.Meanwhile,FBG,fasting insulin,HOMA-IR and leptin were significantly higher in HFD group than in the control group,but the testicular weight and semen parameters were significantly lower.Testosterone level in testis and serum were greatly decreased in HFD group compared with other two groups,while levels of serum and testicular proinflammatory cytokines(TNF-α,IL-6)were elevated.Testicular histology revealed that the diameter of seminiferous tubules and the amount of spermatogenic cells in testicular tissue were greatly reduced in HFD group than that in control and metformin groups.Conclusion:Met can improve the testicular spermatogenic function in obese rats from several aspects,such as improving the glucose and lipidmetabolism,decreasing the disorder of sex hormone secretion,and inhibiting inflammatory reaction of the body and the testis.
