HOXC10高表达与鼻咽癌患者预后的相关性分析

Correlation Analysis Between High Expression of HOXC10 and Prognosis of Nasopharyngeal Carcinoma Patients

目的:探讨HOXC10在鼻咽癌中的表达及其与鼻咽癌患者预后的相关性。方法:采用荧光定量PCR及免疫组织化学的方法对124例无转移的鼻咽癌患者组织中HOXC10 mRNA及蛋白水平表达进行检测。用Chisquare检验对HOXC10的表达与鼻咽癌患者临床病理资料间的相关性进行分析。并采用Kaplan-Meier对HOXC10表达水平与鼻咽癌患者生存率包括疾病特异性生存率(DSS),无转移生存率(MeFS),局部无复发生存率(LRFS)间的相关性进行分析。结果:在鼻咽癌组织中HOXC10表达上调,而HOXC10的高表达与鼻咽癌患者的肿瘤分期(P=0.021)及淋巴结转移(P<0.001)显著相关。单变量分析发现,就DSS(P=0.007)和MeFS(P=0.004 2)而言,HOXC10高表达与鼻咽癌患者差的预后呈正向关。多变量分析发现,HOXC10高表达在预测DSS(P=0.018,风险比=1.988)和MeFS(P=0.026,风险比=1.899)方面仍然是一个独立的预后因子,并且DSS(P=0.026)、MeFS(P=0.033)和LRFS(P=0.014)与鼻咽癌患者的肿瘤分期存在明显的相关性。结论:HOXC10在鼻咽癌中的表达及其与鼻咽癌患者预后存在密切相关性,可能作为鼻咽癌患者预后的一个的生物标志物。

Objective:To evaluate HOXC10 expression and its prognostic effect in a large cohort of nasopharyngeal carcinoma(NPC)patients.Methods:We examined the HOXC10 expression by qRT-PCR and immunohistochemistry in biopsy specimens from 124 patients with non-metastasized NPC.Chi-square analysis was performed on the correlation between HOXC10 expression and clinicopathological variables of NPC patients.The Kaplan-Meier analysis was performed on the correlation between HOXC10 expression and prognosis of patients with nasopharyngeal carcinoma including disease-specific survival(DSS),metastasis-free survival(MeFS),and local recurrence-free survival(LRFS).Results:HOXC10 was up-regulated in NPC tissues.HOXC10 high expression was significantly associated with increment of tumor stage(P=0.021),advanced nodal status(P<0.001).Its expression also correlated with worse prognosis in terms of DSS(P=0.007),MeFS(P=0.004 2)univariately.In multivariate analyses,HOXC10 expression still re-mained prognostically independent to portend worse DSS(P=0.018,hazard ratio=1.978)and MeFS(P=0.036,hazard ratio=1.899),together with stage Ⅲ-Ⅳ(P=0.026,DSS;P=0.033,MeFS).Conclusion:In summary,our results suggest HOXC10 may play a critical role in NPC progression and may serve as a potential prognostic biomarker in NPC patients.
