1型糖尿病大鼠神经重构的实验研究 被引量:2

Experimental Study on Neural Remodeling in Type 1 Diabetic Rat

摘要 目的:采用机械痛阈及病理学方法观察糖尿病大鼠的周围神经病变。方法:SD大鼠25只分为对照组(C组,n=10)和模型组(DM组,n=15)。DM组一次性腹腔注射55mg/kg的链脲佐菌素(STZ)溶液,腹腔注射72h后经尾静脉取血测定空腹血糖,血糖≥16.7mmol/L即为造模成功;C组腹腔注射同等剂量的枸橼酸/枸橼酸钠缓冲液。造模成功后8周测大鼠机械痛阈并观察大鼠腓肠神经超微结构。结果:DM组血糖明显升高且持续整个实验进程;DM组痛阈明显低于C组(P<0.05),腓肠神经出现明显脱髓鞘改变,部分髓鞘溶解消失,同时出现大量新生小神经。结论:糖尿病神经病变时可出现神经重构,髓鞘脱失与再生并存。 Objective:To explore the neural remodeling in type 1 diabetic rats.Methods:Male SD rats were randomly divided into control group and diabetic group.After 8 weeks of streptozotocin(STZ)intraperitoneal injection by 55 mg/kg body weight,the mechanical withdrawal threshold and the morphological changes of sural nerve were observed.Results:Compared with the control group,the mechanical withdrawal threshold was obviously declined in diabetic rats(P<0.05),there was serious demyelination in diabetic rats,some myelin sheath was disappeared,and a large number of new born small nerves appeared.Conclusion:Neural remodeling occurs in diabetic neuropathy,and the demyelination is coexsted with regeneration.
