摘要
目的 探讨甲醛对大鼠外周血细胞DNA的损伤作用 ,研究甲醛的遗传毒性。方法 采用3H -TdR掺入法 ,检测甲醛不同染毒剂量 (1 0、2 0、40mg/kg)及染毒时间 (1 2、2 4、48、72小时 )对大鼠血细胞DNA的损伤情况。结果 在受试剂量、时间范围内 ,3H -TdR掺入量随着染毒剂量增加而明显减少 ,并存在剂量 -反应关系 (r=- 0 80 4 ,P <0 0 1 )。 1 2、2 4、72小时各染毒时间组与对照组差别也有显著性 (P <0 0 5)。结论 甲醛对大鼠外周血细胞DNA具有明显的损伤作用 。
Objective To explore the effect on DNA damage in peripheral blood cells of rats exposed to formaldehyde and study genetic toxicity of formaldehyde.Methods The effect on DNA damage in peripheral blood-cells after i.p.administration of formaldehyde at different dose (10?20?40mg/kg) and for different exposure periods (12?24?48?72 hrs) was measured using 3H-TdR incorporation assay. Results Within tested doses and periods, the extent of DNA damage in blood cells was significantly decreased as the exposure dose increased, showing a dose dependent mannerand ( r =-0 804, P <0 01) and changed significantly for 12?24?72 hrs respectively compared with those of controls( P <0 05).Conclusion Formaldehyde could induce DNA damage of peripheral blood cells of rats. The result indicated once again that formaldehyde had genetic toxicity in character.
