环加氧酶2在人结直肠癌中的表达及其与肿瘤血管形成的关系 被引量:4

Cyclooxygenase-2 expression in human colorectal cancer and its relationship with tumor angiogenesis

摘要 目的探讨环加氧酶2(Cox-2)在结直肠癌中的表达及其与肿瘤血管内皮生长因子(VEGF)表达的关系。方法选取2001年2~9月间在我院手术治疗的35例结直肠癌患者,采用逆转录聚合酶链反应(RT-PCR)的方法,检测肿瘤组织中Cox-2与VEGF的表达,并对结直肠癌Cox-2表达与肿瘤生物学特性的关系进行研究。结果(1)80.0%的肿瘤组织Cox-2表达阳性,31.4%的正常组织表达阳性(P<0.01);(2)肿瘤直径在3~6cm者Cox-2表达率明显高于直径超过6cm者(P=0.0334);(3)高分化组Cox-2表达率较高,而A/B期结直肠癌其表达高于C/D期结直肠癌,但差异均无显著性意义(P>0.05);(4)与Cox-2表达阴性组比较,Cox-2表达阳性组VEGF表达明显增高(25/27),差异有显著性意义(P=0.0030)。结论Cox-2在本组结直肠癌的表达率为80%,且与肿瘤的大小及VEGF表达相关。 Objective To investigate the expression of cyclooxygenase 2(Cox 2) and its relationship with VEGF expression in colorectal cancer. Method RT PCR was used to detect the expression of Cox 2 and VEGF in 35 patients with colorectal cancer during February to September 2001, and correlation of Cox 2 with biological characteristics of colorectal cancer was analyzed. Results (1)80% of the tumor tissues, while 31 4% of normal tissues, were Cox 2 positive (P< 0 01). (2) The positive rate of Cox 2 expression in tumors with a diameter of 3~6 cm was higher than that more than 6 cm (P=0 0334). (3)Cox 2 expression increased in high differentiation tissues as compared to middle and low differentiation tissues, its expression in stage A/B colorectal cancer was higher than that in stage C/D, but no significance was found(P >0 05). (4)VEGF expression in Cox 2 positive group was significantly higher than that in Cox 2 negative group (P=0 0030). Conclusion 80% of colorectal cancer tissues expressed Cox 2 and its expression is correlated with tumor size and VEGF expression.
