十二指肠和小肠肿瘤中血管生成因子的表达及其与微血管计数的关系 被引量:1

Expression of angiogenesis regulatory factors and its relationship with microvessel count in neoplastic tissue of duodenum and small intestine

摘要 目的研究十二指肠和小肠良恶性肿瘤组织中碱性成纤维细胞生长因子(bFGF)信使核糖核酸(mRNA)、血小板源生长因子(PDGF)mRNA和血小板吸附蛋白1(TSP-1)mRNA的表达与微血管计数(MVC)的关系及其临床病理意义。方法MVC染色应用SABC免疫组织化学法,bFGFmRNA、PDGFmRNA和TSP-1mRNA染色为原位杂交染色法。结果腺癌MVC(46.2±7.4)明显高于腺瘤(19.8±8.1)。腺癌bFGFmRNA和PDGFmRNA阳性率(57%,48%)明显高于腺瘤(20%,30%),而TSP-1mRNA则相反(腺癌36%,腺瘤90%)。MVC、bFGFmRNAPDGFmRNA表达阳性高与肿瘤分化差、浸润深、直径大及淋巴结转移有关。腺癌中bFGFmRNA表达与PDGFmRNA表达呈正相关,但与TSP-1mRNA表达呈负相关。结论bFGF、PDGF和TSP-1均为重要的肿瘤血管生成调节因子,其mRNA表达水平与十二指肠和小肠腺癌发生发展、生物学行为及转移的发生有密切关系。 Objectives To study the relationship between expression of bFGFmRNA, PDGFmRNA, TSP 1mRNA and microvessel count (MVC) in neoplastic tissues of duodenum and small intestine,and clarify its clinicopathological significance.Methods The MVC was valued by immunohistochemical method of avidin biotin complex. The expression of bFGFmRNA, PDGFmRNA or TSP 1mRNA were stained by in situ hybridization. Results The MVC was significanfly higher in adenocarcinoma (46.2±7.4) than that in adenoma (19.8±8.1). The possitive rates of bFGFmRNA, PDGFmRNA were significantly higher in adenocarcinoma (57%, 48%) than that in adenoma (20%,30%),which was opposite for TSP 1mRNA. The MVC and the positive rates of bFGF mRNA and PDGF mRNA were significantly higher in the adenocarcinomas with poorer differentiation,deeper infiltration,larger diameter and lymph node metastasis. The expression of bFGF mRNA was correlated positively with that of PDGF mRNA,but negatively with that of TSP 1mRNA. Conclusions bFGF, PDGF and TSP 1 are important regulatary factors of angiogenesis. The expression of their mRNA may be related to the carcinogenesis, progression, biological behaviors, metastatic status of duodenum and small intestinal adenocarcinoma.
