摘要
目的研究骨髓基质中是否具有血液血管干细胞的特征。方法分离并培养人胎儿骨髓基质细胞(hfMSCs),FACS检测细胞的免疫表型。将此细胞接种在基底膜胶中,由血管内皮细胞生长因子(VEGF)和碱性成纤维细胞生长因子(bFGF)诱导,免疫组化和透射电镜检测血管和造血的分化形成。结果此细胞群体的典型特征是CD34阴性的成纤维样细胞99%表达Flk1(即血管内皮细胞生长因子受体2),并能形成血管样结构,而在管样结构形成的过程中诱导出CD34阳性的圆形细胞。结论Flk1+CD34-的hfMSC能向血管内皮细胞和造血细胞分化,提示其具有血液血管干细胞的特征表型。
Objective To investigate whether fetal bone marrow stromal cells have hemang ioblastic charac-teristics.Methods Human fetal bone marrow stromal cells(hfMS Cs)were isolated and cultured.Immunophenotypes of hfMSCs were tested by FACS .hfMSCs seeded in the matrigel were induced with vascular endothelial growth f actor(VEGF)and basic fibroblast growth factor(bFGF)in vitro.Vascularization and hematopoiesis were detected with immunohistochemistry and electron microsco pe.Results The typical properties of this CD34 - stromal cell population were that99%cells expressed Flk1(vascular endot helial cell growth factor receptor2)and tube structure was formed.In the proc ess of induction,hfMSCs could give rise to CD34 + round cells.Conclusions We have demonstrated that fetal bone marrow strom a -derived Flk1 + CD34 - cells could differentiate into vascular endothelial cells and hematopoietic cells,indicating that fetal bone marrow stroma -derived Flk 1 + CD34 - cells have hemangioblastic characteristics.
