沙眼衣原体MOMP基因真核表达载体免疫小鼠的研究 被引量:1

摘要 目的 用pcDNA3-MOMP真核表达质粒直接免疫小鼠 ,观察小鼠对沙眼衣原体MOMP基因疫苗的免疫应答。方法 大量制备重组质粒 pcDNA3-MOMP ,给小鼠肌肉注射 ,每 3周免疫 1次 ,共免疫 3次。用微量免疫荧光及MTT法检测小鼠血清抗体及脾细胞对沙眼衣原体的特异性增殖反应。结果 初次免疫接种后小鼠血清中检测出特异性抗体 ,加强免疫后抗体水平明显上升 ,免疫荧光滴度最高达 1∶2 5 6。脾细胞对沙眼衣原体的刺激指数明显高于对照组 (P <0 0 1)。结论 pcDNA3-MOMP在小鼠体内既可诱导体液免疫应答 ,又可诱导特异性细胞免疫应答。 Aim To study immune responses of DNA vaccination in mice induced by pcDNA3 MOMP Methods The mice were injected intramuscularly with pcDNA3 MOMP and pcDNA3 respectively Each mouse was immunized every three weeks till the third time The specific antibody against C trachomatis in sera of mice was detected by Micro immunnofluoresence (MIF) assay The stimulation index of spleencytes to C trachomatis was measured by MTT Results The significant specific antibody titers were observed after primary immunization After booster immunization,MIF titers in mice increased remarkably up to 1∶256 The stimulation index of spleencytes to C trachomatis were significantly higher than that of mice injected with pcDNA3 Conclusion The investigation demonstrates that pcDNA3 MOMP generated humoral and cellular immune responses in mice
