三倍体山杨组织培养再生系统的建立

Establishment of in Vitro Regeneration System of Triploid Populus Treluma

初步建立了三倍体山杨组织培养再生系统。组织培养时,外植体最好选用健壮生长的顶芽,经HgCl_2溶液表面消毒后,接种到含6-BA0.2mg/L、NAAO.01mg/L的MS培养基上诱导分化。不定芽的增殖最适培养基为含0.05～0.10mg/L的吲哚乙酸的MS培养基。试管苗的生根可采用含0.1mg/L的萘乙酸的1/2MS培养基。移栽成活后,组培苗生长旺盛,整齐一致,表现出良好的生长势。

We have established the tissue culture system of triploid populus tremula. The most suitable ex-plants were shoot-tips. By employing HgC12 solution, surface sterilization of explants was accomplished. The explants were proliferated on MS medium with 6-BA 0.2mg/L, NAA 0. Olmg/L. For in vitro rooting, the shoots were treated on MS medium (half of mi-roelements) with NAA 0. Img/L. The most suitable medium for differentiation was MS medium with 0. 05-0. 1mg/L IAA. After transplantation, the shoots grew vigorously and identically.
