基质金属蛋白酶及其组织抑制物在卵巢癌浸润转移中的作用

摘要 浸润和转移是卵巢癌患者治疗失败和死亡的直接原因,而细胞外基质(ECM)的降解正是卵巢癌浸润正常组织和开始转移的信号及重要途径。目前发现细胞外基质的降解主要依靠4种蛋白水解酶:丝氨酸蛋白酶、半胱氨酸蛋白酶、天冬氨酸蛋白酶及基质金属蛋白酶(MMPs),其中MMPs被认为是最重要的一类,因为:①MMPs直接以酶原的形式分泌到ECM中,并在正常生理条件下发挥作用。②MMPs的表达和活性均受到严格调控。
