摘要
目的 研究 5型腺病毒载体 (Ad5)携带P1 6基因对恶性脑胶质瘤细胞系TJ899生长状态的影响。②方法 免疫组化(SP法 )测定P1 6蛋白表达 ,MTT(methlythiazolyltetrazolium ,MTT)法测定恶性脑胶质瘤细胞系生长状态 ,克隆形成实验。③结果 重组体腺病毒能介导P1 6外源基因在恶性脑胶质瘤细胞系TJ899细胞中阳性表达 ,6d时肿瘤细胞生长抑制率为 93 % ,并且能显著地抑制肿瘤细胞的克隆形成能力。④结论 腺病毒介导P1 6基因能在肿瘤细胞中表达 。
Objective To study the effect of the P16 gene suppressing human glioma cell line TJ899 growth by adenoviral vector(Ad5).Methods P16 expressions was detected in TJ899 cell line by immunohistochemical method.The proliferation of the cell line was analyzed by MTT and colony formation.Results P16 expressions was observed in TJ899 cell line which infected with this recombinant adenovirus.The growth rate of the Ad-P16 infected TJ899 cells was inhibited 93%,compared with that of the Ad-LacZ infected cells.Colony formation in this Ad-P16 infected cells greatly decreased versus Ad-LacZ infected cells.Conclusion The expression of exogenous P16 mediated by adenovirus can inhibit growth and colony formation of infected tumor cells.
