摘要
采用光合菌适用的富集培养基,通过富集驯化的培养方法,从养猪场废水污泥中采集菌液进行较长时间的驯化富集,分离纯化出A2光合菌株,为红色单菌落,革兰氏染色为阴性。对A2菌株生长特性研究表明,添加1.0%Na HCO3能使A2菌株能较早进入生长对数期和稳定期。A2菌株的最适培养条件为:p H为7.5、30℃厌氧光照培养。将菌株A2处理模拟污水和猪场实际污水,探讨菌株A2对氮、磷的去除效果。结果表明,A2菌株处理模拟污水8 d,对TN、NH+4-N、NO-3-N、NO-2-N和TP的平均去除率分别为:74.38%、91.09%、56.17%、90.76%和57.82%。A2菌株处理猪场污水8d,对TN、NH+4-N、NO-3-N、NO-2-N和TP的平均去除率分别为:52.45%、31.19%、45.30%、88.63%和50.92%。表明,菌株A2具有良好的污水处理应用价值。
A high efficiency strain of photosynthetic bacteria named A2 was isolated from pig farm wastewater by using the improved method of enrichment culture medium for a long time. The strain A2 was red single colonies and gram negative staining. Further studied on the growth characteristics of A2 showed that A2 could enter into the logarithmic growth phase and stable phase earlier by putting 1. 0% Na HCO3 in cultivation medium. The optimum cultivate condition for A2 was p H 7. 5、30 ℃ and anaerobic environment under light. When A2 strain treated simulated wastewater after eight days,the average removal rates of TN、NH+4- N、NO-3- N、NO-2- N and TP were 74. 38% 、91. 09% 、56. 17% 、90. 76% and 57. 82%,respectively. When the tests of A2 strain treated piggery wastewater after eight days,the average removal rates of TN、NH+4-N、NO-3-N、NO-2-N and TP were 52. 45% 、31. 19% 、45. 30% 、88. 63% and 50. 92%,respectively. The results showed that the strain A2 had a high quality application value in practical wastewater treatment.
