2016—2017年西安市区公共场所水体军团菌污染情况 被引量:4

Legionella contamination in water bodies of public place in Xi’an in 2016-2017

摘要 目的了解2016—2017年西安市公共场所人工与自然水体中军团菌污染状况。方法将水样处理后进行分离培养和血清学鉴定,并采用荧光定量PCR及环介导等温扩增法(LAMP)进行快速检测。结果共采集水样72份,军团菌检出25份,总阳性率为34.72%;嗜肺军团菌检出9份,占军团菌阳性样品的36.00%,其中血清1型占嗜肺军团菌的66.67%,其他为血清2~15型。荧光定量PCR法显示,军团菌阳性样品33份,阳性率为45.83%,其中嗜肺军团菌16份,占军团菌阳性样品的48.48%;环介导等温扩增技术共检出军团菌阳性样品41份,阳性率为56.94%,其中嗜肺军团菌22份,占军团菌阳性样品的53.66%。两种分子生物学技术的军团菌阳性率高于传统分离培养方法,差异有统计学意义(P<0.05)。结论本次检测的西安市区公共场所水体存在不同程度的嗜肺军团菌污染,应加强日常维护。此外,分子生物学技术对于提高军团菌检出率具有一定价值。 Objective To understand the status of Legionella contamination in artificial and natural water in public places in Xi’an from 2016 to 2017.Methods After the water samples were collected and treated,Legionella was isolated,cultured and identified by serology.Rapid detection was carried out by real-time PCR and loop-mediated isothermal amplification(LAMP).Results A total of 72 water samples were collected,Legionella were detected in 25 samples with the total positive rate of 34.72%.Legionella pneumophila was detected a total of 9 samples,accounting for 36.00%of the positive samples of Legionella,including serotype 1,accounting for 66.67%of Legionella pneumophila,the rest were serotypes 2-15.The results of real-time quantitative PCR showed that 33 samples were positive for Legionella with positive rate of 45.83%,among which 16 cases were positive for Legionella pneumophila,accounting for 48.48%of the positive samples of Legionella;the LAMP showed that 41 samples were positive for Legionella with the positive rate of 56.94%,Legionella pneumophila were found in 22 samples,accounting for 53.66%of the positive samples of Legionella.Conclusion There is Legionella pneumophila contamination to various degrees in artificial water bodies in the investigated public places in Xi’an,and routine maintenance should be strengthened.In addition,molecular biological technology is more effective for improving the detection rate of Legionella.
