血清中丙酮和3种醇类的气相色谱内标同时测定法 被引量:1

Simultaneous determination of acetone and three alcohols in serum by gas chromatography with internal standard

摘要 目的建立使用内标定量同时测定血清中甲醇、乙醇、异丙醇和丙酮的气相色谱法。方法将血清与内标液异丁醇以1∶1混合,以10∶1的分流比进样0.6μl,柱流量1.00 ml/min,采用程序升温,经HP-INNOWAX毛细管柱分离,气相色谱火焰离子化检测器内标法测定。结果在15.70~790.00 mg/L的线性范围内,所得甲醇、乙醇、异丙醇、丙酮的回归方程均呈较好的线性关系,r>0.999。方法的检出限为0.31~0.46 mg/L,定量下限为1.05~1.54 mg/L。批内和批间RSD分别为0.8%~3.6%和2.5%~5.5%,加标回收率为94.4%~101.8%。空气中的10种常见有机物对这4种化合物不造成干扰,样品在-8℃以下可保存14 d。结论该方法操作简单,快速,回收率高,重现性好,适合人群血清中甲醇、乙醇、异丙醇、丙酮的监测。 Objective To establish an internal standard method for simultaneous determination of methanol,ethanol,isopropanol and acetone in serum by gas chromatography.Methods The serum was mixed with isobutanol internal standard solution at ratio of 1∶1,then was injected with split ratio of 10∶1 at column flow rate of 1.00 ml/min.The sample was temperature programmed,separated by HP-INNOWAX capillary column and detected with flame ionization detector by internal standard gas chromatography.Results The separation effect of methanol,ethanol,isopropanol and acetone was good.The linear correlation coefficients of this four compounds were>0.999 5.The limits of detection and the lower limits of quantitation were 0.31-0.46 mg/L and 1.05-1.54 mg/L respectively.The within-run and between-run relative standard deviations were 0.8%-3.6%and 2.5%-5.5%respectively.The recovery rates were 94.4%-101.8%.The ten kinds of common organic compounds in the air did not interfered with the determination of this four chemical compounds.The samples could be preserved at lower than-8℃at least 14 days.Conclusion This method has simple operation and rapid determination,high recovery and strong stability,which is suitable for monitoring methanol,ethanol,isopropanol and acetone in human serum.
