摘要
目的:对灵杆菌素生产工艺进行优化,并对筛选工艺下产品的结构进行初步研究。方法:通过正交实验设计确定灵杆菌素最佳生产条件,并采用理化反应、TLC、HPLC等分析方法对产品的组成结构进行分析。结果:确定灵杆菌素最佳生产条件为:超声破壁时间为30min、苯酚溶液比例为1:30、5倍体积乙醇溶液沉淀。TLC分析结果表明灵杆菌素是由甘露糖、半乳糖、葡萄糖和一种未知糖组成的多聚糖。HPLC分析结果显示灵杆菌素具有明显的多糖特征峰,且其生产质量稳定。结论:科学筛选所确定的生产工艺条件能确保制备的灵杆菌素杂质含量少、质量稳定。
Objective:To optimize the production process and preliminarily study on product structure using the optimization process. Methods:To determine the optimization process conditions by Orthogonal experimental and To analyse product structure by the physical and chemical reaction, TLC,HPLC. Results:The optimization process conditions :the time of ultrasonic wall-breaking is30 min,the ratio of phenol solution is 1:30,the precipitation volume ethanol solution is five times; TLC results show that the Prodigiosin is kind of polysaccharide by mannose,galactose,glucose and unknown sugar;HPLC results show that Prodigiosin presents obvious characteristic peak and production quality is stable. Conclusion: The optimization production process by selecting scientifically can sure less impurity and stable quality on Prodigiosin.
