紫芪方对失血性休克大鼠肠道通透性的影响 被引量:2

摘要 目的观察紫芪方对失血性休克Wistar大鼠肠道通透性的影响。方法实验采用Wistar大鼠,按照改良Wigger’s法复制失血性休克模型,然后将模型大鼠随机分为正常对照组、失血性休克组、紫芪方组、参附组及蒸馏水组(各组n=6)。除正常对照组和休克组,其他组在给予2倍失血量液体复苏后分别灌胃给予不同干预药物,观察肠道黏膜组织结构变化,并测定血浆内毒素含量、肠道细菌移位情况、血浆D-乳酸含量和血浆二胺氧化酶(diamine oxidase,DAO)含量。结果紫芪方组肠黏膜损伤程度明显轻于失血性休克组。紫芪方组血浆内毒素含量、血浆细菌移位菌落数、肝脏细菌移位菌落数和血浆D-乳酸分别为(177.0±45.0)EU/L、(29±8)CFU、(31±16)CFU、(0.39±0.04)mmol/L,较失血性休克组显著降低(P<0.01)。紫芪方组血浆DAO含量为(0.40±0.06)kU/L,较失血性休克组降低(P<0.05),且紫芪方在降低血浆细菌移位菌落数、肝脏细菌移位菌落数和血浆DAO 3个方面的效应强于阳性对照药物参附(P<0.05)。结论紫芪方对失血性休克大鼠肠道通透性有保护作用,为紫芪方保护肠道屏障功能的有效性提供了重要依据。 Objective To observe the influence of Chinese herb medicine( Ziqi) on the intestinal permeability of male Wistar rats with hemorrhagic shock. Methods Hemorrhagic shock model was produced by modified Wigger’s model. Male Wistar rats were divided into five groups randomly( the normal group,the hemorrhagic shock group,the resuscitation and Ziqi compound treatment group,the resuscitation and Shenfu treatment group,the resuscitation and distilled water treatment control group,n = 6). Except for the normal and hemorrhagic shock group,different drugs were given by gastric perfusion after resuscitation with double blood loss of liquid. The structure changes of intestinal mucosa and content of endotoxin in plasma,numbers of translocation of bacteria,content of D-lactic acid and diamine oxidase(DAO) in plasma were evaluated. Results The degree of intestinal injury in Ziqi group was significantly less serious than that in hemorrhagic shock group. Levels of endotoxin in plasma( 177. 0 ± 45. 0) EU / L,numbers of bacteria from plasma( 29± 8) CFU and liver tissues( 31 ± 16) CFU,and levels of D-lactic acid in plasma( 0. 39 ± 0. 04)mmol / L in Ziqi compound group decreased markedly as compared with those in hemorrhagic shock group( P < 0. 01). Levels of DAO in plasma( 0. 40 ± 0. 06) k U / L in Ziqi compound group decreased as compared with those in hemorrhagic shock group( P < 0. 05). The effect of Ziqi compound in decreasing numbers of bacteria from plasma and liver tissues,and levels of DAO in plasma was stronger than the effect of the positive control drug Shen-Fu compound( P < 0. 05). Conclusion Ziqi compound has significant protective effect on maintaining intestinal permeability,which provides an important basis for the protection of the intestinal barrier function by Ziqi compound.
