摘要
目的探讨乳腺癌组织中周期素依赖性激酶10(cyclin-dependent kinase 10,CDK10)基因启动子甲基化状态及其与临床病理参数的联系。方法应用甲基化特异性聚合酶链反应检测CDK10基因启动子在50例乳腺癌组织及相应癌旁组织中的甲基化状态并结合临床病理资料进行分析。结果在乳腺癌组织中CDK10基因甲基化阳性率为60%(30/50),癌旁对照组织未出现CDK10基因甲基化。CDK10基因甲基化同淋巴结转移与临床分期显著相关(P<0.05)。结论 CDK10基因甲基化与乳腺癌临床分期密切相关,可能是参与乳腺癌发展的重要分子事件。
Objective To investigate the methylation status of the cyclin-dependent kinase10(CDK10) gene promoter in breast cancer.Methods Methylation-specific polymerase chain reaction was performed to observe CDK10 gene promoter methyation in 50 cases of breast cancer tissues companied with their adjacent non-cancerous tissues.The relationship between CDK10 gene methyation and clinical pathological parameters were also analyzed.Results Methylated CDK10 gene was frequently observed in 60.0%(30/50) of primary tumors,but not in any normal tissue.CDK10 gene methylation was significantly correlated with lymph node metastasis and TNM staging of tumors(P<0.05).Conclusion CDK10 gene methylation may possess a significant function in the development of breast cancer.
