摘要
为了研究北极狐CBD103基因核心启动子活性区、转录因子结合位点、CBD103基因编码蛋白结构及功能,同时探究该基因的表达调控机制,试验通过RT-PCR扩增及测序技术从北极狐皮肤组织中获得CBD103基因cDNA序列,并利用生物信息学方法对所得序列进行预测分析。结果表明:获得序列全长2 327 bp(包括5’侧翼区2 071 bp、CDs区204 bp、3’侧翼区52 bp),编码67个氨基酸残基。北极狐CBD103基因5’侧翼区存在潜在的启动子区域和CpG岛,且发现GC框、TATA框、CAAT框等调控元件及Sp1、Ets、Mef-2、ERE、Myc等多种转录因子。CBD103基因编码的蛋白是定位于细胞核和线粒体的不稳定疏水性蛋白质,此蛋白无规卷曲所占比例较大,使得蛋白构象多样化,主要参与信号转导和转录,存在跨膜区域和信号肽。北极狐与家犬的遗传亲缘关系最近。说明CBD103基因在不同物种间具有一定的保守性,且各物种间亲缘关系与生物进化观点基本一致,同时符合动物分类学。
The aim of the present study was toidentify the Arctic fox CBD103 gene core promoter activity domain,transcription factor binding sites,structure and function of CBD103 protein,and to explore the expression regulation mechanism of the gene. The c DNA sequences of CBD103 gene was obtained from Arctic fox skin tissue by PCR amplification and sequencing. And the bioinformatics methods were used to predict and analyze the resulting sequences.The results showed that the total length of the c DNA sequence was 2 327 bp( including 2071 bp 5’ flanking region,204 bp CDs region and 52 bp 3’ flanking region).The gene encoded 67 amino acid residues.There was a potential promoter region and a Cp G islandin the Arctic fox CBD103 gene. Some regulatory elementthes( such as TATA box,CAAT box and GC box) and transcription factors( such as Sp1,Ets,Mef-2,ERE and Myc) were also found in the CBD103 gene. The protein encoded by CBD103 was an unstable hydrophobic protein which was located in the nucleus and mitochondria.This protein had a large proportion of random coil,which might lead to a variety of protein conformation. The protein might play a role in signal transduction and transcription. The transmembrane region and signal peptide were also found in the protein. The genetic relationship between the Arctic fox and the Canis lupus familiaris was the most recent. The results indicated that CBD103 gene was a conserved protein among different species. And the genetic relationship between different species was consistent with the biological evolutionand animal taxonomy.
