摘要
为了对山羊子宫内膜上皮细胞不同发情时期MUC1基因与蛋白水平的表达趋势进行分析,试验采用浓度为1×10^(-5 )mg/mL孕酮、1×10^(-7 )mg/mL雌二醇培养子宫内膜上皮细胞48 h,利用激光共聚焦显微技术、荧光定量PCR技术及蛋白免疫印迹法分析比较MUC1的表达差异。结果表明:MUC1蛋白主要在子宫内膜上皮细胞的细胞质中表达;MUC1 mRNA相对表达量呈先上升后下降的趋势;MUC1蛋白相对表达量呈先上升后下降的趋势,且发情中期MUC1蛋白相对表达量较高,发情间期MUC1蛋白相对表达量最低(P<0.05)。说明在不同发情时期山羊子宫内膜上皮细胞中MUC1的转录翻译水平变化存在规律,且主要受类固醇激素影响。
In this experiment,uterine endometrial epithelial cells were cultured with 1×10-5mg/mL progesterone and 1×10-7mg/mL estradiol for 48 h.The expressions of MUC1 gene were detected and compared by laser confocal microscopy,qPCR,and Western-blot.The results show that MUC1 protein is mainly expressed in the cytoplasm of endometrial epithelial cells.The mRNA relative expressions of MUC1 gene show a trend of increasing first and then decreasing.The relative expressions of MUC1 protein show a trend of increasing first and then decreasing.In the middle estrus period,the relative expression of MUC1 protein is relatively high.The relative expression of MUC1 protein in the estrous period is the lowest(P<0.05).It indicates that the transcription and translation levels of MUC1 in goat endometrial epithelial cells in different estrus cycles are regulated and are mainly affected by steroid hormones.
