胰岛素/油酸钠复合物的制备及鉴别

摘要 制备胰岛素/油酸钠复合物,对其制备工艺进行考察,提高胰岛素在水中的表观油水分配系数,以期进行进一步的制剂研究。采用非水离子对法制备胰岛素/油酸钠复合物,并以单因素考察优化制备工艺,对复合物进行红外分析,考察胰岛素表观油水分配系数的变化。胰岛素/油酸钠复合物的制备工艺为:油酸钠与胰岛素摩尔比为6∶1,室温下磁力搅拌(500rpm)0.5h。按照此方法成功制备了胰岛素/油酸钠复合物,复合物在水中的油水分布系数得到明显的提高。 To improve apparent partition coefficient by preparing Insulin/sodium oleate(Insulin/S.O)complex.Insulin/S.O complex was prepared by hydrophobic ion pairing(HIP)method.The effects of many factors including molar ratio of S.O to insulin,reaction temperature,reaction time and rate of rotation were studied to get optimized process,then the apparent partition coefficient of insulin and FITR analysis were studied to confirm the formation of Insulin/S.O complex.The optimized preparation conditions were determined as follows:the molar ratio of S.O to insulin,the reaction temperature,the rate of rotation and the reaction time were 6∶1,ambient temperature,500 rpm and 0.5h,respectively.Insulin/S.O complex can be made by HIP method and the apparent partition coefficient of insulin can be evidently improved by the method.
