白藜芦醇对肺动脉高压大鼠肺组织的抗氧化损伤作用 被引量:4

Effect of Resveratrol on Anti-oxidative Injury of Lungs in Rats with Pulmonary Arterial Hypertension

摘要 目的研究白藜芦醇对肺动脉高压大鼠肺组织抗氧化损伤作用。方法 48只雄性SD大鼠随机分为4组:对照组、野百合碱组、白藜芦醇组、伐地那非组。对照组给予2 ml生理盐水腹腔注射,其余三组同时给予野百合碱腹腔注射造成肺动脉高压模型,正常饲养21 d。而后各组给予相应药物处理21 d后测量肺动脉平均压(mean pulmonary artery pressure,mPAP)、右心肥厚指数RV/(LV+S),处死动物,留取左肺组织,比色法检测肺组织匀浆上清液中的超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽(glutathione,GSH)、丙二醛(malondialdehyde,MDA)、过氧化氢酶(catalase,CAT)的表达。结果同对照组相比,其余三组mPAP、RV/(LV+S)显著增高(P<0.05);同野百合碱组相比,白藜芦醇组、伐地那非组mPAP、RV/(LV+S)显著下降(P<0.05)。同野百合碱组相比,白藜芦醇、伐地那非治疗后能够明显降低大鼠肺组织MDA含量(P<0.05),增加SOD、GSH、CAT的含量(P<0.05)。结论大鼠肺动脉高压造模后予白藜芦醇处理,可有效减轻肺动脉高压大鼠肺组织氧化应激损伤,对肺动脉高压肺组织氧化应激损伤有一定逆转作用。 Objective To investigate the anti-oxidative injury effects of resvertrol on lungs of MCT-induced pulmonary arterial hypertension rats.Methods A total of 48 male Sprague-Dawley(SD)rats were randomly and equally devided into 4 groups.Control group was injected with normal saline(NS),and the others with Monocrotaline(MCT).All animals were fed for 21 days normally.Control group was injected with NS,the other three groups were treated with NS(MCT group),Resvertrol(Res group)and Vardenafil(Val group)for 21 to 42 days respectively.The mean pulmonary artery pressure(mPAP),right ventricular hypertrophy index(RV/(LV+S))were measured and compared among all groups.Then animals were sacrificed,and superoxide dismutase(SOD),glutathione(GSH),malondialdehyde(MDA),catalase(CAT)expression in the left lung tissue were detected.Results Compared with control group,mPAP and RV/(LV+S)were significantly increased in the other three groups(P<0.05).Compared with MCT group,decreased the producing of MDA,improved expression levels of SOD,GSH and CAT were observed in rats treated with Res or Var(P<0.05).Conclusion Treatment of MCT-induced PAH rats with resveratrol can attenuate reactive oxygen species injury.Resveratrol exerts antioxidant effect in lung tissue,which may contribute to reversal oxidative stress injury.
