IL-18基因启动子区-137C/G和-607C/A多态性与大动脉炎发病的相关性

Correlation between IL-18 Promoter Region-137C/G and-607C/A Polymorphism with Arteritis

目的探讨白细胞介素18(interleukin 18,IL-18)基因启动子区-137C/G和-607C/A多态性与大动脉炎发病的相关性。方法选取2008-03/2014-04月在作者医院治疗的大动脉炎患者104例(病例组),同时选取相匹配的104例健康者(对照组),分别对两组进行基因分型,分析-137C/G和-607C/A位点的等位基因和基因型的分布情况。结果 IL-18基因启动子区-607C/A位点基因型分别为AA型、CC型和AC型,IL-18基因启动子区-137C/G检测到3种基因型:CC型、GC型和GG型;IL-18基因-137C/G和-607C/A等位基因和基因型频率符合Hardy-Weinberg平衡;在未校正危险因素之前,-137C/G和-607C/A在显性模式、加性模式和隐性模式下的比值比分别为1.150和0.720、1.500和0.716、1.709和0.833,校正年龄、性别等危险因素后,在显性模式、加性模式和隐性模式下的比值比分别为1.073和0.673、1.423和0.684、1.673和0.783。结论人群中IL-18基因启动子区-137C/G和-607C/A存在多态性,但大动脉炎发病与IL-18基因启动子区-137C/G和-607C/A单核苷酸多态性相关性不强。

Objective To study the relationship between interleukin 18(IL-18) gene promoter region-137C/G,-607C/A polymorphism and arteritis.Methods From 2008 March to 2014 April,104 patients with arteritis in our hospital(treatment group) and 104 matching healthy subjects(control group) were studied.The distribution of-137 C / G and-607 C / A allele and the genotypes of the two groups were analysed.Results The promoter regions of IL-18 gene-607 C / A locus genotypes were AA type,CC type and AC type.The promoter regions of IL-18 gene-137 C / G were detected in CC type,GC type and GG type.IL-18 gene-137 C / G and-607 C / A allele and genotype frequencies conformed with Hardy-Weinberg equilibrium.Before adjustment for risk factors,the odds ratios(OR) of-137 C / G and-607 C / A in the dominant model,additive model and recessive model were1.150 and 0.720,1.500 and 0.716,1.709 and 0.833 respectivley.After correciton for age,sex and other risk factors,the OR in the dominant model,additive model and recessive model were 1.073 and 0.673,1.423 and 0.684,1.673 and 0.783 respectivley.Conclusion The crowd IL-18 gene promote gene-137 C / G and-607 C / A polymorphism exists,but the correlation between arteritis pathogenesis and IL-18 gene promoter-137 C / G and-607 C / A single nucleotide polymorphism is not strong.
