丹参酮ⅡA减轻野百合碱诱导的大鼠肺动脉高压模型IL-6和IL-8的表达 被引量:4

摘要 目的建立野百合碱诱导的肺动脉高压的大鼠模型,观察丹参酮ⅡA对大鼠血清中的白细胞介素6(interleukin 6,IL-6)和IL-8的表达的影响,探讨丹参酮ⅡA对防治肺动脉高压的意义。方法 SD大鼠分为野百合碱组、丹参酮ⅡA治疗组及对照组,野百合碱组给予大鼠肌肉注射野百合碱制成肺动脉高压大鼠模型,丹参酮ⅡA治疗组在注射野百合碱的基础上给予注射丹参酮ⅡA,对照组不作处理。分别测定3组右心室收缩压(right ventricular systolic pressure,RVSP)、平均肺动脉压力(mean pulmonary arterial hypertension,mPAP)、右心室肥厚指数(right ventricle hypertrophy index,RVHI)。用酶联免疫法(enzyme linked immunosorbent assays,ELISA)检测血清中的IL-6和IL-8的含量。结果成功建立了野百合碱诱导的肺动脉高压大鼠模型,使用丹参酮ⅡA可以减低野百合碱引起的肺动脉压力的升高及IL-6和IL-8的表达。结论 IL-6及IL-8参与了野百合碱诱导的肺动脉高压,丹参酮ⅡA可以降低野百合碱诱导的肺动脉高压,可能与减轻肺动脉高压的炎症反应有关。 Objective To establish the pulmonary hypertension induced by monocrotaline in rats model,observation of tanshinoneⅡA in the serum of rat interleukin6(interleukin6,IL-6)and the influence of the expression of IL-8,discussing the meaning of tanshinoneⅡA for prevention and treatment of pulmonary hypertension.Methods SD rats were divided into monocrotaline group,tanshinone ⅡA treatment group and control group.Monocrotaline group was given monocrotaline,tanshinone ⅡA treatment group was given monocrotaline and tanshinone ⅡA,control group without making processing.Three groups were determined right ventricular systolic pressure(RVSP),mean pulmonary arterial hypertension(mPAP),right ventricle hypertrophy index(RVHI).IL- 6 and IL 8 in serum were detected by enzyme league immunoassay(ELISA).Results Successfully established the pulmonary hypertension induced by monocrotaline in rats model.Using tanshinone type IIA could reduce pulmonary arterial pressure caused by monocrotaline and the expression of IL-6 and IL-8.Conclusion IL-6 and IL-8 are involved in the pulmonary hypertension induced by monocrotaline.Tanshinone IIA can reduce the process,it may be to reduce the inflammation associated with pulmonary hypertension.
