桑青枯雷尔氏菌MR111的gsp I和gsp J基因的克隆与序列分析

摘要 gspI和gsp J两个蛋白可能参与青枯雷尔氏菌II型分泌系统周质复合体的形成。为进一步研究青枯雷尔氏菌基因间的互作,以桑青枯雷尔氏菌MR111为材料,对MR111的gsp I和gsp J基因进行了PCR扩增和测序,分别获得了gsp I基因(391 bp)和gsp J基因(567 bp)序列;序列编码蛋白均含典型的该类基因的功能性结构,N端均以疏水氨基酸亮氨酸(L)和丙氨酸(A)最多,具分泌型信号肽特性,与其他青枯雷尔氏菌的同源基因的一致性在94%以上;以gsp I和gsp J基因联合矩阵进行聚类分析,发现几种青枯雷尔氏菌基因聚合后,再与其他雷尔氏菌属基因相聚合。 The bacterial wilt ( Ralstonia solanacearum ) is one of the major diseases in mulberry. Two proteins gsp I and gsp J may be involved in the formation of periplasmic complex of Ralstonia solanacearum type II secretion system. The genes gsp I and gsp J of strain MR111 with 391 bp and 567 bp, respectively, have been obtained by PCR amplication and sequence. The sequence-encoded protein contains functional structures of this gene type with the secretory signal peptide composed mainly by leucine and alanine in the N-end of the two genes. The identity between these two genes and other homologous genes from R. solanacearum is both above 94%. The results of clustering analysis with gsp I and gsp J gene combined matrix show that all genes from R. solanacearum have been clustered in one branch, and then would be clustered with the genes from Ralstonia sp. and R. Pickettii in this study.
