高粱辐射诱变早熟品种(系)的同工酶分析

ANALYSIS OF ISOZYMES FROM THE SORGHUM EARLY CULTUVARS(STRAINS)INDUCED BY IRRADIATION

摘要 用聚丙烯酰胺凝胶电泳对高粱3个经^(60)Coγ20kR 辐射育成的品种(系)和3个对照品种进行了过氧化物酶同工酶、酯酶同工酶分析,结果表明:(1)经^(60)Coγ20kR 处理育成的3个生育期短的品种(系)与对照品种的两种同工酶比较,在芽早期辐育品种(系)的酶谱带数及酶活性均高于对照品种,芽后期辐育品种(系)的过氧化物酶同工酶谱带数及酶活性低于对照品种,酯酶同工酶则趋于一致。(2)各品种总酶谱是一致的。由此推论,用传统的"基因突变"解释经辐射育成的新品种(系)是值得进一步商榷的。 Isozymes of peroxidase and esterase from the 3 early cultivars(strains) sorghum induced by irradiation and the 3 check strains were analysed by using polylamide gel electrophoresis (PAGE).The results revealed: (1) By comparing isozymes of peroxidase and esterase of 3 short-de- velopment-stage cultivars (strains) induced by ^(60)Co γ-ray irradiation with the checks,the bands on PAGE and enzyme activity of the cultivars (strains)were more and higiher than those of the checks in early sprou- ting.The bands on PAGE activity of peroxidase isozyme of the cultivars (strains)were less and lower than those of the checks,but the zymogram of esterase isozyme of cultivars(strains)and that of the checks appeared to be similar in later sprouting. (2) In spite of the enzyme diversity at one growth stage between diffe- rent cultivars,the overall enzyme patterns of all the growth stages were simi- lar to each other,which presented a great similarity to the essential pattern reported. It is worth while discussing that the cultivars (strains) induced by ra- diation were explained by tradional"gene mutation"theory.
