摘要
目的 :观察紫花牡荆素(CAS)是否能抑制HeLa细胞系宫颈癌球形成细胞即宫颈癌干细胞样细胞(CCSLCs)自我更新能力。方法:体外培养HeLa细胞系细胞。干细胞条件培养基悬浮培养得到和扩增CCSLCs。MTT法测定细胞活性,肿瘤球形成法检测自我更新能力,Western blot分析磷酸化Akt蛋白表达。结果:CAS优先抑制CCSLCs细胞活性,同时,显著抑制CCSLCs自我更新能力,呈浓度依赖性(P<0.05)。与亲本HeLa细胞系细胞比较,CCSLCs细胞磷酸化Akt表达水平增高。CAS有效地降低CCSLCs细胞磷酸化Akt蛋白表达水平。结论:CAS抑制源自HeLa细胞系CCSLCs自我更新作用与其下调细胞磷酸化Akt蛋白表达有关。
Objective To observe whether casticin(CAS) inhibits the self-renewal capacity of sphere-forming cells derived from cervical cancer HeLa cell line, namely, cervical cancer stem-like cells (CCSLCs). Methods Hela cell line was cultured in vitro. CCSLCs were obtained and amplified by suspended culture method with stem cell-conditioned medium. The viability of cells was examined by the MTT assay and the capacity of Self-renewal was detected by tumorsphere-forming assay. The protein expression level of p-AKT was analyzed using western blot. Results CAS preferentially inhibited the viability of cell proliferation in CCSLCs, and significantly sup-pressed their capacity of self-renewal, in a concentration-dependent manner (P<0.05). The express level of p-AKT was elevated in CCSLCs, compared with the parental cells. CAS effectively down-regulated the expression level of p-AKT protein in CCSLCs. Conclusion CAS inhibits self-renewing capacity of CCSLCs derived from cervical cancer HeLa cell line cells is associated with down-regulation of p-AKT expression.
