摘要
目的:检测哮喘小鼠小气道及肺组织IL-21、MMP-2与TIMP-1的表达及布地奈德干预对其表达的影响;分析IL-21与MMP-2、TIMP-1及MMP-2/TIMP-1比值的相关关系。方法:建立哮喘小鼠模型。36只雌性小鼠随机分为3组,每组12只。分别为生理盐水对照组(CTRL组)、卵清蛋白(ovalbumin,OVA)致敏、激发哮喘组(OVA组)、布地奈德干预组(OVA+BUD组)。观察各组小气道及肺组织HE及Masson染色情况;免疫组化检测MMP-2、TIMP-1蛋白的表达;RT-PCR检测IL-21的表达。结果:HE及Masson染色结果示哮喘小鼠模型制作成功;OVA组MMP-2与TIMP-1的表达、MMP-2/TIMP-1比值高于CTRL组与OVA+BUD组,其差异有统计学意义(P<0.05);CTRL组与OVA+BUD组比较,其差异无统计学意义(P>0.05);OVA组IL-21的表达低于CTRL组与OVA+BUD组,其差异有统计学意义(P<0.05);CTRL组高于OVA+BUD组,其差异有统计学意义(P<0.05)。结论:OVA组IL-21的表达低于CTRL组和OVA+BUD组;OVA组MMP-2、TIMP-1的表达及MMP-2/TIMP-1比值高于CTRL组和OVA+BUD组。布地奈德干预后,IL-21表达上调,MMP-2、TIMP-1的表达与MMP-2/TIMP-1比值降低IL-21与MMP-2、TIMP-1的表达及MMP-2/TIMP-1比值呈负相关。
Objective To observe the expressions of Interleukin 21 (IL-21), matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of matrix metallo-proteinases-1 (TIMP-1) in lung tissue of asthma mice and the ef-fect of budesonide on the expression;To analyse the relationship between the IL-21 and MMP-2 ,TIMP-1 as well as MMP-2/TIMP-1 ratio. Methods Mice model of asthma was established. Thirty-six female mice were randomly divided into 3 groups, with twelve in each group. The first group was the normal saline control group (CTRL group), the second group was OVA sensitization and activation of asthma group (OVA group) and the third group was the budesonide intervention group (OVA+BUD group). Observe the lung tissue in each group of mice for HE staining and Masson staining;Conduct immunohistochemical detection for the expressions of MMP-2 and TIMP-1 protein;Conduct RT-PCR detection for the expressions of IL-21. Results 1.HE and Masson staining of lung tis-sue paraffin section show the asthma mice model has been successfully established. 2.The expressions of MMP-2 and TIMP-1 as well as MMP-2/TIMP-1 ratio in OVA group are higher than those of CTRL group and CTRL+BUD group, where the difference is statistically significant (P<0.05);To compare the expressions of MMP-2 and TIMP-1 as well as MMP-2/TIMP-1 ratio in both the CTRL group and OVA+BUD group, the difference is not sig-nificant (P>0.05). 3.The expression level of IL-21 in the small airway and lung tissue in OVA group is lower than that of CTRL group and OVA+BUD group, the difference is statistically significant (P<0.05);the expression level of IL-21 in CTRL group is higher than that of OVA+BUD group, the difference is statistically significant (P<0.05).Conclusion 1.The expression of IL-21 in the lung tissue of asthma mice is lower than that of mice in normal saline control group and budesonide intervention group;The expression of MMP-2 and TIMP-1 and MMP-2/TIMP-1 ratio of asthma mice are higher than those of CTRL group and CTRL+BUD group. 2.After the interven-tion of budesonide, the expression of IL-21 in the lung tissue of asthma mice is up-regulated, and the expression of MMP-2 and TIMP-1 and MMP-2/TIMP-1 ratio is down-regulated. 3.Both the expressions of IL-21 and MMP-2 and TIMP-1 and MMP-2/TIMP-1 ratio are negatively correlated.
