摘要
目的:延长离断肢体的活性保存时间,为未来战时后方离体组织复合移植手术创造条件,促进再植肢体功能恢复,减少致残率。方法:将40只Wistar大鼠随机分为4组,形成离断肢体模型后,分别采用UW液、HTK液、林格氏液+地塞米松+低分子肝素钠及生理盐水在4℃冰箱内进行24小时持续灌注。收集不同时间点的灌出液及组织病理标本,观察生化指标和光镜下组织结构病理变化。结果:对大鼠离断肢体模型进行持续灌注后,灌注组与对照组相比较,相同时间点发生病理损伤程度轻,生化指标存在统计学差异(P<0.05)。UW液灌注组与HTK液及林格氏液+地塞米松+低分子肝素钠灌注组相比较,发生病理损伤程度轻,生化指标与其余组相比无统计学差异(P>0.05)。结论:对离断肢体进行灌注保护可以有效的减缓组织损伤,延长组织活性。在不同灌注液之中使用UW液灌注,可以使离断肢体得到更好的保护效果。
Objective To extend the preservation time of the amputated limbs which keep activity and create the conditions for future transplant operations that may happen in war times. Promote the functional recovery of the amputated limbs after operations and reduce the rate of disability. Methods 40 Wistar rats were randomly divided into 4 groups,and then produced the models of amputated limbs of rats,The four groups were treated with UW solution,HTK solution,Ringer solution + Dexamethasone + low molecular solution and Physiological saline severally. All of the four groups were bored continuously in 24 hours in the refrigerator at 4 ℃,then collected the irrigation fluids of different times and Pathological samples to detect the biochemical indexes and observe the changes of organizational structure under light microscope. Results After being treated with different perfusates,the amputated limb had lighter changes of organizational structure than those were treated with Physiological saline,and there was significant difference in biochemical index.( P < 0. 05) Among three perfusates,UW solution had the lightest changes of organizational structure,and there was no significant difference in biochemical index.( P > 0. 05) Conclusion It had protective effects on the amputated limbs which were treated with perfusates,and the UW solution had stronger protective effects on the amputated limbs than any other perfusates.
