摘要
目的:采用HPLC法比较板蓝根药材及其农本方颗粒中表告依春的含量。方法:采用反相高效液相色谱法,色谱柱为Kromail C18(250mm×4.6mm,5μm);以乙腈-水(15∶85)为流动相,流速:1.0 mL/min;检测波长245 nm,柱温30℃。结果:表告依春在6.3~84.0μg/mL范围内浓度与峰面积成良好的线性关系,r=0.9995;板蓝根粉末平均回收率为99.13%,RSD为1.21%;农本方颗粒平均回收率为99.09%,RSD为1.31%;测得板蓝根粉末以及农本方颗粒中表告依春含量分别为4.81mg/g,4.53mg/g。结论:本实验建立的高效液相色谱法准确、可靠,可用于板蓝根药材及其制剂的质量评价。结果表明板蓝根农本方制剂中的表告依春含量达到其药材中含量的90%以上。农本方颗粒制剂值得推广。
Objective To compare the content of epi-goitrinin in Radix Isatidis and its NONG's granule determining by high performance liquid chromatography method. Methods The HPLC was carried out using a Kromasil C18( 250 ×4.6 mm ID,5μm) analytical column at 30 ℃,with a mixture mobile phase[Acetontrile: water( 15∶85) =86∶14]at the flow rate of 1mL/minwith the wavelength at 263 nm. Results The linear range of epigoitrinin is 6. 3 ~ 84. 0 μg /mL,r = 0. 9995; The mean recovery of epi-goitrinin in Radix Isatidis and its NONG's granule is 99. 13% ± 1. 21% and 99. 09 ± 1. 31%,respectively. Conclusion The high performance chromatography developed in this investigation is accurate and credible,which is suitable for the assay of epi-goitrinin in Radix Isatidis and its NONG's granule. The results showed that the content of epi-goitrinin if NONG's granule is arrived more than 90% compared with that of Radix Isatidis,which was the base for the application of NONG's granule.
