摘要
目的 :研究大肠癌组织及癌旁正常组织PTPRO基因启动子甲基化状态及m RNA表达情况,并探讨其在大肠癌发生中的关系。方法 :应用甲基化特异性PCR(MSP)和RT-PCR分别检测30例大肠癌组织及癌旁正常组织中PTPRO基因启动子甲基化状态及m RNA表达状态。结果 :大肠癌组织中PTPRO基因启动子甲基化率高于癌旁正常组织,其甲基化样本中PTPRO基因m RNA表达明显受抑。结论 :PTPRO基因启动子甲基化在大肠癌发生中起着重要作用,可能与大肠癌的发生、发展及预后有关。
Objective Research the PTPRO Gene promoter methylation and the expression of PTPRO gene m RNA in colorectal cancer tissue and normal tissue adjacent to carcinoma,and to discuss their relationship in colorectal cancer. Methods Methylation specific PCR(MSP) and RT-PCR were use to detected PTPRO gene promoter methylation status and m RNA’s expression in 30 cases of colorectal cancer tissue and in normal tissue adjacent to carcinoma. Results The PTPRO gene promoter methylation rate of colorectal cancer tissues is higher than normal tissue adjacent to carcinoma,the PTPRO gene m RNA’s expression of the methylation samples is obviously inhibitive. Conclusion The PTPRO gene promoter methylation plays an important role in colorectal cancer, and it may be related to the development and prognosis of colorectal cancer
