苯扎贝特对小鼠肝脏SR-B1的影响

摘要 目的 :检测苯扎贝特干预后小鼠肝脏SR-B1m RNA表达,探讨苯扎贝特干预后对小鼠肝脏B族I型清道夫受体(SR-B1)m RNA的影响。方法 :35只C57BL/6小鼠随机分为5组,分别给予普通饲料、不同剂量的苯扎贝特(0.1%、0.25%、0.5%、0.75%)添加普通饲料喂养4周后,应用逆转录聚合酶链反应测定小鼠肝脏SR-B1m RNA表达。结果 :不同剂量苯扎贝特(0.1%、0.25%、0.5%、0.75%)干预后小鼠肝脏SR-BIm RNA的表达分别为0.22±0.07、0.53±0.04、0.84±0.01、0.92±0.05,与对照组0.11±0.04比较,苯扎贝特明显增加小鼠肝脏SR-BIm RNA的表达。结论 :苯扎贝特能够上调肝脏SR-B1m RNA表达,促进体内胆固醇逆转运,利于动脉粥样硬化防治。 Objective Aim This study is to observe effect of benzafibrate on The Scavenger receptor class B type I.(SR-BI) mR NA expression of liver. Methods 35 male C57BL/6 mice were randomly divided into four groups and treated with ordinary diet or plus different dosage(0.1%, 0.25%, 0.5%, 0.75% W/W) of bezafibrate for 4 weeks. Reverse transcription polymerase chain reaction(RT-PCR) was used to evaluate mR NA expression level of SR-BI. Results The SR-BImR NA expression of liver in mice treated with bezafibrate(0.1%, 0.25%, 0.5%, 0.75%) were upregulated by 0.22±0.07、0.53±0.04、0.84±0.01、0.92±0.05,compared to those in control group. Conclusions Bezafibrate upregulate the mR NA expression of SR-B1 to mediate the selective uptake of cholesteryl esters from HDL in the liver, so it will be in favor of the prevention of atherosclerosis.
