摘要
目的 :研究血浆mi R-126、mi R-143在急性心肌梗死AMI和不稳定型心绞痛UAP、稳定型心绞痛(SAP)及非冠心病患者(NCHD)中浓度的变化。方法 :收集急性心肌梗死22例和不稳定型心绞痛31例,稳定型心绞痛患者(23例)及非冠心病患者(20例)共96例,采集入院后24小时内的清晨空腹静脉血标本,以mi R-39为参照基因,Trizol法提取血浆总micro RNA,逆转录成c DNA,并用real-time q PCR检测血浆中micro RNA的相对表达量,分析micro RNA相对表达量各组中的变化。结果 :1.mi R-126:与NCHD组比较,SAP组表达量增高,且有统计学差异,AMI、UAP组有增高趋势,但无统计学意义;2.mi R-143:与NCHD组比较,UAP、SAP组无差异,AMI组明显升高;AMI组高于UAP和SAP组,且具有统计学差异;结论 :1.mi R-126在SAP组较AMI、UAP、NCHD均明显升高,且均有统计学差异,说明有可能在AS发病过程中具有保护作用。2.mi R-143在AMI中高表达,且与UAP、SAP、NCHD相比均有统计学差异,表明mi R-143在某种程度上有可能作为AMI时的临床检测指标之一。
Objective To investigate the expression of peripheral plasma miR-126, miR-143 in patients with coronary artery disease(CAD) and NCHD. Methods We collected the fasting venous blood samples of 96cases, with acute myocardial infarction 22cases, 31cases of unstable angina pectoris,23cases of stable angina pectoris(SAP)and 20 cases of non-coronary heart disease (NCHD), within 24 hours since their admission. We measured the relative expression of microRNA in their plasma by real-time polymerase chain reaction (PCR), and analysed the microRNA parameters in four groups. Results 1.miR-126 ex-pression in SAP is higher than NCHD group, with statistical significance; the expression in AMI and UAP is higher than NCHD group,but with no statistical significance. 2.miR-143 expression in AMI group was significantly higher than UAP,SAP and NCHD groups, with statistical significance;there were no significant difference among UAP ,SAPand NCHD groups,with no statistical significance Conclusions 1.miR-126 may be a protection factor in AS,it would be a novel method to delay or prevent AS.2.miR-143 expression in plasma may be a another novel biomarker for AMI.
