摘要
目的:建立简单方便的测定黄嘌呤氧化酶的方法。方法:在现有方法的基础上,采用十二烷基硫酸钠(SDS)为终止剂,改进硝基四氮唑蓝法测定了黄嘌呤氧化酶的方法。SDS浓度为10%。酶反应时间为25min。结果:与原方法相比,改进的方法具有不产生蓝色沉淀、不必离心就可直接测定,准确度、精密度好的优点。适合于测定在紫外区有吸收的化合物对黄嘌呤氧化酶的作用。结论:以SDS为终止剂的硝基四氮唑蓝法简单易行,适合于黄嘌呤氧化酶抑制剂的筛选。
Objective To establish a simple and convenient method for determination of xanthine oxidase(XOD). MethodsOn the basis of previous research, an improved method applied with nitroblue tetrazolium was used for the determination of the xanthine oxidase with 10% sodium dodecyl sulfate(SDS) as terminating agent. The reaction time for the enzyme was 25 min. Results The activity of XOD can be determined with improved method which can directly determined without centrifugation since no blue precipitation produced, it is more accuracy and precise compared with the original method. This method is suitable for the investigation on the effect of compounds with ultraviolet absorption on XOD. Conclusion The modified method with SDS as terminator is simple and suitable for the screening of xanthine oxidase inhibitors.
