摘要
目的:探究miR-586在骨肉瘤患者体内的表达及意义。方法:选取我院2014年9月~2016年9月收治的骨肉瘤患者151例,将其作为观察组,选择同期于我院进行体检的健康的志愿者52例,将其作为对照组,对患者瘤体周围正常骨组织及骨肉瘤瘤体组织中miR-586的表达、两组血清miR-586的表达、患者血清中miR-586表达与骨肉瘤患者瘤体组织的相关性、miR-586对骨肉瘤的诊断价值进行分析。结果:瘤体周围正常骨组织、骨肉瘤瘤体组织的miR-586相对表达水平分别为(0.94±0.27)、(5.34±0.29),患者瘤体周围正常骨组织较骨肉瘤瘤体组织中miR-586相对表达水平高,差异明显具有统计学意义;观察组、对照组血清miR-586的相对表达水平分别为(1.48±0.47)、(0.85±0.22),观察组较对照组高,差异明显,具有统计学意义;经皮尔森相关系数分析发现,患者血清中miR-586与骨肉瘤患者瘤体组织呈正相关的关系;经对观察组、对照组患者外周血miR-586水平ROC曲线分析,曲线下面积为0.882(0.774、0.991为95%可信区间;P=0.023),当0.9551为最佳筛查阳性届值(cut-off)时,特异度及敏感度均为85%。结论:miR-586对骨肉瘤患者具有较好的诊断价值,其表达水平可作为骨肉瘤患者诊断的生物学指标。
Objective To investigate the expression and significance of miR-586 in osteosarcoma patients. Methods 151 patients with osteosarcoma in our hospital from September 2014 to September2016 were selected as the observation group. In the same period, 52 healthy volunteers were selected as our control group. Tumor surrounding normal bone tissue and osteosarcoma tissues and the expression of miR-586, two groups of patients with serum miR-586 expression in serum of patients with miR-586 and patients with osteosarcoma tumor correlation, the diagnostic value of miR-586 on osteosarcoma were analyzed. Results The relative expression levels of miR-586 in the normal bone tissue and osteosarcoma tissue around the tumor were(0.94+0.27),(5.34+0.29). The relative expression of miR-586 in tumor tissues was higher than that in osteosarcoma tissues, and the difference was statistically significant. The relative expression levels of serum miR-586 in the observation group and the control group were(1.48+0.47),(0.85+0.22). The observation group was higher than the control group, and the difference was statistically significant. There was a positive correlation between miR-586 in serum and tumor tissue in osteosarcoma patients by Pearson correlation coefficient analysis. The miR-586 level and ROC curve of peripheral blood in the observation group and the control group were analyzed, the area under the curve was 0.882(0.774, 0.991 is 95% confidence interval; P=0.023), and when 0.9551 was the best screening positive term value(cut-off), the specificity and sensitivity were 85%. ConclusionMi R-586 has better diagnostic value for osteosarcoma, and its expression level can be used as a biological marker for the diagnosis of osteosarcoma.
