摘要
目的:观察TW37对食管癌TE-1细胞迁移的影响。方法:用0. 25、0. 50、1. 00、2. 00、4. 00、8. 00、16. 00、32. 00和64. 00μmol/L的TW37处理TE-1细胞24、48、72 h,采用MTT法检测细胞增殖。倒置显微镜下观察2、4μmol/L的TW37作用24 h对TE-1细胞形态的影响。划痕实验检测2、4μmol/L的TW37作用24、48 h对TE-1细胞迁移的影响。Western blot实验测定2、4μmol/L TW37作用24 h对TE-1细胞E-Cadherin、Vimentin以及N-Cadherin蛋白表达的影响。均设空白对照。结果:TW37抑制TE-1细胞增殖,呈时间和浓度依赖性。TW37作用后TE-1细胞由原来的梭形变成了卵圆形。TW37能够在较低浓度(2μmol/L)抑制TE-1细胞迁移,4μmol/L的TW37抑制迁移的作用更强(P <0. 05)。TW37能降低TE-1细胞中Vimentin、N-Cadherin的表达,上调E-Cadherin的表达(P <0. 05)。结论:TW37可能通过抑制EMT进程进而抑制TE-1细胞的迁移。
To observe the effects and possible mechanism of TW37 on the migration of esophageal cancer TE-1 cells. Methods: TE-1 cells were treated with 0.25,0.50,1 00,2 00,4.00,8.00, 16.00, 32.00, and 64. 00 μmol /L TW37 for 24, 48, and 72 h,respectively,and MTT assay was used to detect cell proliferation. TE-1 cells were treated by 2,4 μmol /L TW37 for 24 h,the morphology of TE-1 cells was recorded by inverted microscope,and the expressions of E-Cadherin, Vimentin and N-Cadherin were measured by Western blot. TE-1 cells were treated by 2,4 μmol /L TW37 for 24, 48 h, respectively,and cell migration was detected by wound healing assay. Results: TW37 significantly inhibited the proliferation of TE-1 cells in a time-and concentration-dependent manner. TE-1 cells changed from the original shuttle into an ovalshape after being treated by TW37. TW37 could inhibit the migration of TE-1 cells at a lower concentration,which was stronger with higher concentration( P < 0. 05) . Western blot results confirmed that TW37 could significantly reduce the expressions of Vimentin and N-Cadherin, and up-regulate that of E-Cadherin( P < 0. 05) . Conclusion: TW37 might inhibit the migration of TE-1 cells by regulating the expressions of EMT related proteins.
