大鼠H9c2心肌细胞中SK2通道和JP2蛋白的表达及定位

摘要 目的:研究大鼠H9c2心肌细胞中SK2通道和JP2蛋白的表达及定位,探讨二者的相互作用。方法:采用Western blot检测SK2通道及JP2蛋白在H9c2细胞中的表达,免疫共沉淀鉴定JP2与SK2通道之间的相互作用,免疫荧光标记观察SK2通道和JP2蛋白在H9c2细胞的定位。结果:心肌细胞H9c2表达SK2通道及JP2蛋白,SK2通道和JP2蛋白可形成免疫复合物,SK2通道和JP2蛋白存在部分共定位。结论:心肌细胞H9c2中SK2通道与JP2蛋白存在相互作用。 Aim:To investigate the expression and localization of SK2 channel and JP2 protein in rat H9c2 cardiomyocytes,which provides the evidence underlying the relationship between SK2 channel and JP2 protein.Methods:Western blot was used to detect the expressions of the SK2 channel and JP2 protein in the rat H9c2 cardiomyocytes.Both co-immunoprecipitation and immunofluorescence techniques were applied to determine the interaction and sub-localization of SK2 and JP2 in the cells.Results:The SK2 channel as well as JP2 protein were expressed in the rat H9c2 cardiomyocytes.The immune complex of SK2 channel and JP2 protein existed in the cells,and they were partly co-located in the cells.Conclusion:The interaction of SK2 channel with JP2 protein exists in the rat H9c2 cardiomyocytes.
