摘要
目的:探讨PolyI∶C复合物对小鼠体内非小细胞肺癌(NSCLC)移植瘤的作用。方法:将C57BL/6小鼠随机分为4组,每组6只,均皮下注射LL/2细胞建立NSCLC移植瘤模型。当肿瘤体积达到100 mm3时,滴鼻组每隔1天PolyI∶C滴鼻1次(0.3 mg/次),肌注组每隔1天肌内注射PolyI∶C 1次(0.3 mg/次),PBS组每隔1天PBS滴鼻1次(100μL/次),PD1组腹腔注射PD1(100μg/次,1周1次)。用药2周后处死小鼠,剥离移植瘤及脏器组织,TUNEL法检测移植瘤组织内细胞凋亡,HE染色观察各脏器肿瘤转移情况。结果:给药后4组小鼠体重均逐渐增加,但肌注组小鼠体重增加幅度小于其他3组(P<0.05)。滴鼻组和肌注组移植瘤体积较PBS组减小(P<0.05),而肌注组减小更为显著(P<0.05)。滴鼻组、肌注组和PD1组抑瘤率分别为(38.33±0.05)%、(50.39±0.03)%、(15.17±0.05)%,肌注组抑瘤率最高(P<0.05)。滴鼻组、肌注组及PD1组肿瘤细胞凋亡均较PBS组增强,且肌注组变化最为显著(P<0.05)。肌注组小鼠各脏器均未发生转移。结论:PolyI∶C复合物对小鼠体内NSCLC移植瘤具有明显的抗肿瘤、抗转移作用。
Aim:To investigate the effects of PolyI∶C complex on transplanted tumor of non-small cell lung carcinoma(NSCLC) in mice.Methods:C57 BL/6 mice were injected subcutaneously with LL/2 cells into the right flank to establish transplantation model of NSCLC,then were randomly allocated into 4 groups,and 6 in each group.When the average tumor volume reached 100 mm3,the treatment was started.Intranasalness group was administered intranasally PolyI∶ C complex(0.3 mg per time,every other day),injection group was intramuscully injected PolyI∶ C complex(0.3 mg per time,every other day),PBS group was administered intranasally PBS(100 μL per time,every other day),and PD1 group was intraperitoneally injected PD1(100 μg per time,1 time per week).After two weeks,the transplanted tumor and visceral organs were dissected,TUNEL method was used to detect cell apoptosis,and HE staining was used to observe the metastasis.Results:The weight of the mice in the 4 groups increased gradually from administration,and the increase range of the intranasalness group was less than those of other 3 groups(P<0.05).The volume of transplanted tumors in the intranasalness group and the injection group was less than that of the PBS group,the change of the injection group was more obviously(P<0.05).The tumor inhibition rate of the intranasalness group,injection group and PD1 group was(38.33±0.05) %,(50.39±0.03) %,(15.17±0.05) %,and that of the injection group was the highest(P<0.05).Cell apoptosis of the intranasalness group,injection group and PD1 group was enhanced,and the change of the injection group was more obviously(P<0.05).There was no metastasis in visceral organs in the injection group.Conclusion:PolyI∶ C complex has obvious anti-tumor and anti-metastatic effects on NSCLC transplanted tumor in mice.
