摘要
目的:探讨ZFAND3基因rs9470794多态性与河南农村汉族人群2型糖尿病易感性的关系。方法:采用1∶1匹配病例对照设计,从河南农村队列研究中分别抽取526对2型糖尿病患者和对照,采用SNPscanTM高通量SNP分型技术进行基因分型;应用条件logistic回归模型分析rs9470794位点多态性与2型糖尿病易感性的关系。应用logistic回归和非线性混合效应模型探讨基因-环境交互作用。结果:病例组和对照组rs9470794基因型分布频率差异有统计学意义(P=0. 026);调整混杂因素后,突变纯合子(CC)携带者发生2型糖尿病的风险是非携带者的1. 750倍(95%CI=1. 075~2. 848)。交互作用分析显示,rs9470794基因型与BMI对2型糖尿病的发生存在相乘交互作用(OR=1. 304,95%CI=1. 199~1. 419)。结论:ZFAND3基因rs9470794多态性与2型糖尿病易感性有关,且该位点多态性与BMI对2型糖尿病的发生存在相乘交互作用。
Aim: To explore the association of zinc finger AN1-type containing 3( ZFAND3) gene rs9470794 polymorphism with the risk of type 2 diabetes mellitus( T2 DM) in a rural Han population in Henan. Methods: Based on the 1∶ 1 matched case-control design,a total of 526 pairs of patients with T2 DM and controls were collected from the Henan Rural Cohort Study. The SNPscanTMKit was used to detect the genotypes. Conditional logistic regression was used to evaluate the association of rs9470794 with the risk of T2 DM. Logistic regression and nonlinear mixed effect model were used to estimate the gene-environment interactions. Results: The distribution of rs9470794 genotypes frequency had significant difference between the two groups( P = 0. 026). Compared with non-carriers,the carriers of mutation homozygote( CC) were 1. 750 times more likely to develop T2 DM after adjusting confounding factors( OR = 1. 750,95% CI = 1. 075-2. 848). The multiplicative interaction between rs9470794 polymorphism and BMI was detected( OR = 1. 304,95% CI = 1. 199-1. 419).Conclusion: ZFAND3 gene rs9470794 polymorphism is associated with T2 DM,and there is multiplicative interaction between rs9470794 and BMI in the risk of T2 DM.
