平溃灌肠剂对溃疡性结肠炎模型大鼠疗效及作用机制研究

摘要 目的:研究平溃灌肠剂对溃疡性结肠炎大鼠模型的治疗作用,并对其作用机制进行探讨。方法:选取SPF级SD大鼠60只,随机抽取8只作为正常组,剩余52只采用三硝基苯磺酸+乙醇制备溃疡性结肠炎大鼠模型。造模成功后剩余50只大鼠随机分为模型组、柳氮磺吡啶组(0.32g/kg)及平溃灌肠剂高(17.25g/kg)、中(8.64g/kg)、低(4.32 g/kg)剂量组,每组10只。给予相应药物进行治疗,连续给药10 d。观察大鼠病变活动指数(DAI)变化,进行损伤程度评分、病理组织学评分,检测血清D-乳酸(D-LA)、二胺氧化酶(DAO)水平及脱氧胆酸(UDCA)、牛磺熊脱氧胆酸(TUDCA)、甘氨熊脱氧胆酸(GUDCA)水平。结果:各给药组大鼠体质量均显著高于模型组(P<0.05);各给药组DAI、结肠黏膜组织损伤程度评分、病理组织学评分均显著低于模型组(P<0.05);各给药组与模型组比较,表现为D-LA、DAO和DCA降低,UDCA、TUDCA和GUDCA升高,其中柳氮磺吡啶组、平溃灌肠剂高剂量组各指标与模型组比较,差异均有统计学意义(P<0.05)。结论:平溃灌肠剂可能通过促进溃疡性结肠炎大鼠结肠黏膜损伤修复,降低D-LA、DAO和DCA,升高UDCA、TUDCA和GUDCA水平发挥其治疗作用。 Objective:To study the therapeutic effect of Pingkui enema on rat model of ul cerative colitis and to explore its mechanism.Methods:A total of 8 SPF SD rats were randomly selected as the normal group,and the remaining 52 rats were treated with trinitrobenzenesulfonic acid+ethanol to establish ulcerative colitis model.After successful modeling,the remaining 50 rats were randomly divided into model group,salazosulfapyridine(SASP)group(0.32 g/kg)and Pingkui enema high-dose group(17.25 g/kg),medium-dose group(8.64 g/kg)and low-dose group(4.32 g/kg)dose group,with 10 rats in each group.The corresponding drug was given for treatment for 10 days.The changes of pathological activity index(DAI)in rats were observed,the degree of injury score and histopathological score were made,and the levels of serum D-lactic acid(D-LA),diamine oxidase(DAO),deoxycholic acid(UDCA),taurous deoxycholic acid(TUDCA)and glycine ursodeoxycholic acid(GUDCA)were detected.Results:The body weight of rats in each administration group was significantly higher than that of model group(P<0.05).The DAI,damage degree score and histopathological score of colonic mucosa in each administration group were significantly lower than those in model group(P<0.05).Compared with the model group,D-LA,DAO and DCA were decreased in each administration group,and UDCA,TUDCA and GUDCA were increased.Among them,there were statistically significant differences between the SASP group,Pingkui enema high-dose group and the model group.Conclusion:Pingkui enema may play a therapeutic role in promoting the repair of colonic mucosal injury in rats with ulcerative colitis,reducing D-LA,DAO and DCA,and increasing the levels of UDCA,TUDCA and GUDCA.
