活血解毒方对干燥综合征小鼠的干预机制 被引量:8

摘要 目的:探讨活血解毒方对干燥综合征小鼠血清白细胞介素-6(interleukin,IL-6)、IL-21及颌下腺病理的影响及机制。方法:以NOD/Ltj小鼠及C57bl/6j小鼠为实验对象,分为正常组、模型组、白芍总苷组、活血解毒方组。正常组、模型组分别给予蒸馏水灌胃,另外两组分别给予白芍总苷混悬液、活血解毒方汤剂灌胃,干预8周。比较各组小鼠唾液流率、颌下腺指数、颌下腺病理染色及评分,并收集血清检测干预后各组小鼠血清IL-6、IL-21水平。结果:正常组唾液流率均高于其他各组(P<0.05);在第8周时,活血解毒方组唾液流率高于模型组及白芍总苷组(P<0.05)。正常组颌下腺指数均高于其余各组(P<0.05)。干燥综合征小鼠各组颌下腺病理评分均高于正常组(P<0.01);与模型组比较,活血解毒方组评分较低(P<0.05)。模型组IL-6水平高于正常组,而白芍总苷组及活血解毒方组血清IL-6水平低于模型组(P<0.05);模型组及白芍总苷组血清IL-21水平高于正常组(P<0.01);活血解毒方组血清IL-21水平低于模型组及白芍总苷组(P<0.05)。结论:活血解毒方可改善干燥综合征模型小鼠唾液流率,减轻小鼠颌下腺炎症,降低血清IL-6、IL-21水平。 Objective:To investigate the effect and mechanism of Huoxue Jiedu Decoction on serum interleukin(IL-6),IL-21 and submandibular gland in mice with Sj?gren′s syndrome.Methods:NOD/Ltj mice and C57 bl/6 j mice were divided into normal group,model group,and the total glucosides of Radix Paeoniae Alba as well as Huoxue Jiedufang group.The normal group and the model group were given distilled water respectively,and the other two groups were given the total glucosides of Radix Paeoniae Alba and Huoxue Jiedu Fang Decoction for 8 weeks.Serum flow rate,submandibular gland index,submandibular gland pathological staining and scoring were compared between the groups,and serum IL-6 and IL-21 levels were measured after serum intervention.RESULTS:The salivary flow rate of the normal group was higher than that of the other groups(P<0.05).At the 8 th week,the salivary flow rate of the Huoxue Jiedu Recipe group was higher than that of the model group and the total glucosides of paeony(P<0.05).The submandibular gland index of the normal group was higher than the other groups(P<0.05).The pathological scores of submandibular glands in the mice with Sj?gren′s syndrome were higher than those in the normal group(P<0.01).Compared with the model group,the scores of Huoxue Jiedu Recipe group were lower(P<0.05).The IL-6 level in the model group was higher than that in the normal group,and the serum IL-6 level was lower in the total glucosides and the Huoxue Jiedu decoction group than in the model group(P<0.05).The serum IL-21 level of the model group and total glucosides of paeony was higher in the group than in the normal group(P<0.01).The serum IL-21 level in the Huoxue Jiedu formula group was lower than that in the model group and the total glucosides of paeony(P<0.05).Conclusion:Huoxue Jiedu Recipe can improve the salivary flow rate of mice with Sjogren′s syndrome,reduce the inflammation of submandibular glands in mice,and reduce the levels of serum IL-6 and IL-21.
