葛花总黄酮对脑缺血再灌注模型小鼠死亡率及生化指标的影响 被引量:4

Effects of Total Flavonoids from Pueraria Lobata on Mortality and Biochemical Parameters in Mice with Cerebral Ischemia-reperfusion

摘要 目的:通过观察葛花总黄酮对小鼠脑缺血再灌注模型的影响,从而探讨葛花总黄酮对脑缺血再灌注损伤的保护机制。方法:小鼠反复脑缺血再灌注模型采用双侧颈总动脉阻断血流10 min,恢复灌流10 min,再次阻断10 min的方法。将小鼠分为7组,阳性药组、葛花总黄酮各剂量组分别连续灌服相应药物7 d,每天1次,假手术组、模型组给予同体积生理盐水。最后1次给药1 h后手术造模,24 h后取脑组织,测小鼠脑组织中炎症因子白细胞介素-6(interleukin-6,IL-6)、白细胞介素-10(interleukin-10,IL-10)、肿瘤坏死因子-α(Tumor Necrosis Factor-α,TNF-α)水平;检测血清神经元特异性烯醇化酶(neuron-specific enolase,NSE)水平。结果:小鼠反复脑缺血再灌注模型造模成功,葛花总黄酮各剂量组可降低小鼠死亡率,降低模型小鼠脑组织中IL-6、TNF-α水平和血清NSE水平,升高脑组织中IL-10水平。结论:葛花总黄酮可改善脑组织炎症反应,减少神经细胞凋亡,增加神经细胞营养因子保护神经元,减少小鼠脑缺血反复再灌注模型损伤,起到对脑组织的保护作用。 Objective:To investigate the protective mechanism of total flavonoids from Pueraria lobata on cerebral ischemia-reperfusion injury in mice.Methods:The repeated cerebral ischemia-reperfusion model in mice was performed by blocking the blood flow of the common carotid artery for 10 min,restoring perfusion for 10 min,and blocking again for 10 min.The mice were divided into 7 groups,the positive drug group and the total flavonoids of Pueraria lobata.Each dose group was continuously administered with the corresponding drug for 7 days,once a day,and the sham operation group and the model group were given the same volume of normal saline.After the last administration for 1 hour,the model was established.After 24 hours,the brain tissue was taken.The inflammatory factor I interleukin-6(IL-6)and interleukin-10(interleukin-10)were measured in the brain tissue of the mouse.IL-10),Tumor Necrosis Factor-α(TNF-α)levels;serum neuron-specific enolase(NSE)levels were measured.Results:The model of repeated cerebral ischemia-reperfusion in mice was successful.The total flavonoids of Pueraria lobata could reduce the mortality of mice and reduce the levels of IL-6,TNF-αand serum NSE in the brain tissue of model mice.IL-10 levels in high brain tissue.Conclusion:The total flavonoids of Pueraria lobata can improve the inflammatory response of brain tissue,reduce the apoptosis of nerve cells,increase the neurotrophic factors to protect neurons,reduce the damage of cerebral ischemia and reperfusion model in mice,and protect the brain tissue.
