补肾舒脊颗粒提取物免疫抑制作用研究

Study on Immunosuppressive Effect of Extract of Bushen Shuji Granule

目的:开展补肾舒脊颗粒提取物对免疫系统调节作用的研究,阐明药物的作用特点。方法:采用Ⅱ型胶原诱导的DBA/1小鼠免疫性关节炎模型,观察补肾舒脊颗粒提取物对关节炎模型小鼠关节炎指数评分、脾脏湿质量和血清中抗Ⅱ型胶原抗体含量的影响。通过绵羊红细胞所致小鼠溶血素抗体生成模型,观察补肾舒脊颗粒提取物对绵羊红细胞所致小鼠特异性抗体生成的影响。结果:补肾舒脊颗粒提取物给药8 d、14 d、18 d、23 d、28 d后关节炎指数评分较模型组显著降低,且能够显著降低模型动物脾脏湿质量和血清抗Ⅱ型胶原抗体IgG的表达。补肾舒脊颗粒提取物能够降低绵羊红细胞所致小鼠特异性抗体的生成。结论:补肾舒脊颗粒提取物具有较好的治疗免疫性关节炎和体液免疫抑制的作用,提示免疫抑制作用可能是本药效应机制的重要环节。

Objective:To study the effect of Bushen Shuji granule extract on immune system and clarify the characteristics of drug action.Methods:The effects of Bushen Shuji granule extract on arthritis index score,spleen wet quality and serum anti-type II collagen antibody content were observed in DBA/1 mice induced by type II collagen.The effect of Bushen Shuji granule extract on the production of specific antibody in mice induced by sheep erythrocyte through the production model of hemolysin antibody in mice induced by sheep erythrocyte was observed.Results:After 8,14,18,23 and 28 days of administration,the arthritis index score of Bushen Shuji Granule extract was significantly lower than that of model group,and the wet quality of spleen and the expression of anti-collagen II IgG in serum of model animals were significantly reduced.The extract of Bushen Shuji Granule can reduce the production of specific antibodies in mice induced by sheep red blood cells.Conclusion:The extract of Bushen Shuji Granule has good therapeutic effects on immune arthritis and humoral immunosuppression,suggesting that immunosuppression may be an important part of the mechanism of the drug.
